Detrimental effect of Bisphenol S in mouse germ cell cyst breakdown and primordial follicle assembly

چکیده

The female reproductive lifespan is largely determined by the size of primordial follicle pool, which established in early life. Bisphenol S (BPS), frequently present plastic products used daily life, has been demonstrated as an exogenous estrogen-like endocrine disrupting chemical interfering with and systems. However, molecular mechanisms its toxicity remain to be determined. In study, we focused on effect BPS ovarian folliculogenesis mice. Our vivo experiments showed that treatment at 2 10 ?g/kg body weight/day for 3 days induced abnormal germ cell cyst breakdown assembly mouse ovary, further affecting later differentiation reducing oocyte quality. addition, our vitro study could interact estrogen receptors (ERs) induce phosphorylation JNKs, responsible adhesion cysts. Meanwhile, exposure up-regulated Notch signaling pathway increase proliferation granulosa cells precursors. provided new evidence adverse effects reproduction, especially after perinatal exposure, elucidated how it works.

Follistatin288 Regulates Germ Cell Cyst Breakdown and Primordial Follicle Assembly in the Mouse Ovary

In mammals, the primordial follicle pool represents the entire reproductive potential of a female. The transforming growth factor-β (TGF-β) family member activin (ACT) contributes to folliculogenesis, although the exact mechanism is not known. The role of FST288, the strongest ACT-neutralizing isoform of follistatin (FST), during cyst breakdown and primordial follicle formation in the fetal mic...

The effect of BMP4 on mouse embryonic stem cell proliferation and differentiation into primordial germ cells

Background and Aim: Artificial gamete production from stem cells is a novel strategy for treatment of infertility. Among various stem cell sources, embryonic stem cells (ESC) can be considered as an appropriate source for in vitro formation of germ cells. In this study we evaluated the effect of BMP4 on proliferation and differentiation of mouse embryonic stem cells into primordial germ cells (...

Estrogen can signal through multiple pathways to regulate oocyte cyst breakdown and primordial follicle assembly in the neonatal mouse ovary.

During mouse embryonic development, oocytes develop in germline cysts, formed by several rounds of cell division followed by incomplete cytokinesis. Shortly after birth, cysts break down and individual oocytes are enclosed by granulosa cells to form primordial follicles. At the same time, two-thirds of the oocytes die by apoptosis with only one-third surviving. We have previously shown that the...

Insulin regulates primordial-follicle assembly in vitro by affecting germ-cell apoptosis and elevating oestrogen.

Insulin is a protein secreted by pancreatic β-cells, which plays an important role in the regulation of ovarian function. However, the specific molecular mechanism of its function remains largely unknown. This study aimed to assess the effect of insulin on mouse folliculogenesis using an in vitro ovary-culture model. The results demonstrated that insulin promoted the proliferation of ovarian gr...

Cell-cell interactions in primordial follicle assembly and development.

Two critical processes in ovarian biology are the assembly of the primordial follicles early in development and then the subsequent development and transition of the primordial follicle to the primary follicle. These processes directly effect the number of oocytes available to a female throughout her reproductive life. Once the pool of primordial follicles is depleted a series of physiological ...
