Intravitreal Administration of Retinal Organoids-Derived Exosomes Alleviates Photoreceptor Degeneration in Royal College of Surgeons Rats by Targeting the Mitogen-Activated Protein Kinase Pathway

چکیده

Increasing evidence suggests that exosomes are involved in retinal cell degeneration, including their insufficient release; hence, they have become important indicators of retinopathies. The exosomal microRNA (miRNA), particular, play roles regulating ocular and functions, photoreceptor maturation, maintenance, visual function. Here, we generated organoids (ROs) from human induced pluripotent stem cells differentiated a conditioned medium for 60 days, after which were extracted ROs (Exo-ROs). Subsequently, intravitreally injected the Exo-RO solution into eyes Royal College Surgeons (RCS) rats. Intravitreal administration reduced apoptosis, prevented outer nuclear layer thinning, preserved function RCS RNA sequencing miRNA profiling showed miRNAs mainly mitogen-activated protein kinase (MAPK) signaling pathway. In addition, expression MAPK-related genes proteins was significantly decreased Exo-RO-treated group. These results suggest Exo-ROs may be potentially novel strategy delaying degeneration by targeting MAPK

Inactivation of mitogen-activated protein kinase signaling pathway reduces caspase-14 expression in impaired keratinocytes

Objective(s):Several investigations have revealed that caspase-14 is responsible for the epidermal differentiation and cornification, as well as the regulation of moisturizing effect. However, the precise regulation mechanism is still not clear. This study was aimed to investigate the expression of caspase-14 in filaggrin-deficient normal human epidermal keratinocytes (NHEKs) and to explore the...

Targeting the RAS pathway by mitogen-activated protein kinase inhibitors.

Targeting of oncogenic driver mutations with small-molecule inhibitors resulted in powerful treatment options for cancer patients in recent years. The RAS (rat sarcoma) pathway is among the most frequently mutated pathways in human cancer. Whereas targeting mutant Kirsten RAS (KRAS) remains difficult, mutant B rapidly accelerated fibrosarcoma (BRAF) kinase is an established drug target in cance...

Photoreceptor rescue of pigment epithelium-derived factor-impregnated nanoparticles in Royal College of Surgeons rats

PURPOSE To investigate the protective effect of intravitreal injection of pigment epithelium-derived factor-impregnated nanoparticles (PEDF-NPs) against photoreceptor degeneration in Royal College of Surgeons (RCS) rats. METHODS Three-week-old RCS rats received an intravitreal injection of PBS, blank NPs, PEDF (2.5 μg), or PEDF-NPs (2.5 μg). Eyes were assessed with morphological, immunohistoc...

Changes in glutamate homeostasis cause retinal degeneration in Royal College of Surgeons rats.

The aim of the present study was to investigate glutamate homeostasis in retinal degeneration-induced changes and the potential mechanisms of glutamate-mediated excitotoxicity in a rat model. The expression of vesicular glutamate transporter-1 (VGLUT-1) and protein kinase Cα (PKCα) in wild-type and Royal College of Surgeons (RCS) rat retinas, at postnatal Day 15 (P15), P30, P60 and P90, were de...

Preservation of inner retinal responses in the aged Royal College of Surgeons rat. Evidence against glutamate excitotoxicity in photoreceptor degeneration.

PURPOSE The aged Royal College of Surgeons (RCS) rat with advanced retinal degeneration loses the b-wave and shows a negative-going corneal electroretinogram (ERG) that has been attributed to loss of inner retinal function because of glutamate toxicity. The authors investigated the origin of this negative ERG and evaluated inner retinal function in late-stage RCS degeneration. METHODS The ERG...
