Concerning Histidine and Carnosine

چکیده

Our knowledge of carnosine is largely due to the admirable investigations of Gulewitsch. The discovery of the base in beef muscle extract was first reported by Gulewitsch and Amiradiib? in 1900. Later Kutsche? isolated a substance from beef muscle extract, which he named ignotine, but Gulewitscl? showed that ignotine and carnosine were identical. In the same volume, he4 showed that hydrolysis of the base with barium hydroxide liberated the amino-acid, histidine. 5 years later, he5 established the identity of the other component, namely @-alanine. This aminoacid had been found in beef extract by Engeland,‘j as a result of the hydrolysis of carnosine during the process of isolation. Thus 11 years after its discovery, Gulewitsch showed that carnosine consisted of but two amino-acids, probably joined to form a dipeptide. Either one of two linkages was possible. Thus far no conclusive evidence had been advanced in favor of either formula. On the whole little has been done to establish the relative distribution, biological significance, and fate of carnosine in the

Histidine and carnosine delay diabetic deterioration in mice and protect human low density lipoprotein against oxidation and glycation.

In vivo effects of histidine and carnosine against diabetic deterioration in diabetic Balb/cA mice were studied. Histidine and carnosine at 0.5, 1 g/l were added into drinking water. After 4 weeks intake of these agents, the content of histidine and carnosine in plasma, heart and liver significantly elevated (P < 0.05). The intake of these agents significantly decreased plasma glucose and fibro...

Interactions between carnosine and captopril on free radical scavenging activity and angiotensin-converting enzyme activity in vitro.

Interactions between carnosine (beta-alanyl-L-histidine), being plentiful in skeletal muscles and neuronal tissues, and captopril, a widely used angiotensin-converting enzyme (ACE) inhibitor, were examined concerning free radical scavenging activity and ACE activity in vitro. Not only captopril, but also carnosine, at concentrations less than those ordinarily found in muscles and neuronal tissu...

Impairment of jejunal absorption rate of carnosine by glycylglycine in man in vivo.

Using a double-lumen tube jejunal perfusion system in vivo, the mutual effects of carnosine (beta-alanyl-L-histidine) and glycylglycine on their respective absorption rates have been studied in six Zambian African adults. Data on the effect of the constituent amino-acids of carnosine on glycylglycine absorption rate have similarly been obtained. The solutions infused in each subject contained (...

Biosynthesis, release, and uptake of carnosine in primary cultures.

Biosynthesis, release, and uptake of carnosine (beta-alanyl-L-histidine) in highly enriched primary cell cultures of skeletal muscle and CNS tissue have been investigated. The synthesis is restricted to muscle cells, oligodendrocytes, and ensheathing cells of olfactory bulb and increases during differentiation of these cells. Astrocytes, in contrast, do not synthesize carnosine but are equipped...

Concentrations of carnosine, anserine, L-histidine and 3-methyl histidine in boar spermatozoa and sheep milk by a modified HPLC method.

The present study deals with the application of high-performance-liquid-chromatography (HPLC) method for a quantitative detection of carnosine, anserine, L-histidine and 3-methyl-L-histidine in biological material with o-phthaldialdehyde (OPA) post column derivatisation at the constant temperature of 50 degrees C. For this purpose, some mobile-phases were prepared with scalar acetonitrile conce...
