چکیده

This paper reviews the molecular and cellular events during precartilage condensation and describes the application of a new retroviral gene transduction procedure. In micromass cultures, dissociated mesenchymal cells interact to form cartilage nodules. Cells first form small aggregates, then bigger clusters. Next, through rearrangement, they consolidate to form precartilage condensations which then differentiate into cartilage. We mapped molecular expression in these early stages and showed that the sequence of molecular expression was tenascin-C, cyclic adenosine monophosphate (cAMP) response element binding protein (CREB), and phos-phorylated CREB (PCREB) → neural cell adhesion molecules → integrin β1 and fibronectin → collagen II and Alcian blue. We then used a newly developed replication competent avian sarcoma (RCAS) mediated gene transduction procedure to alter the gene expression in micromass cultures. We showed that RCAS-(antisense NCAM) can suppress cartilage formation by forming smaller and fewer cartilage nodules. Sonic hedgehog directs mesenchymal cells to form novel nodules with characteristics of hypertrophic cartilage. The use of retroviral transduction, together with the addition of growth factors and Fab’ antibodies to the media, constitute powerful approaches to the analysis of the molecular cascade of chondrogenesis in micromass cultures. It also provides a means to engineer the fate of mesenchymal cells through modulation of adhesion molecules and transgene expression of signaling molecules.

In vitro cartilage formation by human adult stem cells from bone marrow stroma defines the sequence of cellular and molecular events during chondrogenesis.

One approach to resolving the complexities of chondrogenesis is to examine simplified systems in vitro. We analyzed cartilage differentiation by human adult stem cells from bone marrow stroma. Marrow stromal cells were cultured as micromass pellets for 21 days in serum-free medium containing transforming growth factor (TGF)-beta3, dexamethasone, and bone morphogenetic protein (BMP)-6. Assays fo...

Reduced COX-2 Expression in Aged Mice Is Associated With Impaired Fracture Healing

The cellular and molecular events responsible for reduced fracture healing with aging are unknown. Cyclooxygenase 2 (COX-2), the inducible regulator of prostaglandin E(2) (PGE(2)) synthesis, is critical for normal bone repair. A femoral fracture repair model was used in mice at either 7-9 or 52-56 wk of age, and healing was evaluated by imaging, histology, and gene expression studies. Aging was...

Platelet-derived growth factor A modulates limb chondrogenesis both in vivo and in vitro

Cartilage formation in the chick limb follows rapid proliferation, condensation and differentiation of limb mesenchyme. The control of these early events is poorly understood. Platelet-derived growth factor receptor alpha (PDGFR-alpha) is present throughout the mesenchyme of early chick limb buds, while its ligand, PDGF-A, is expressed in the surrounding epithelium. PDGFR-alpha is down-regulate...

The effect of high frequency electric field on enhancement of chondrogenesis in human adipose-derived stem cells

Objective(s):Osteoarthritis (OA) is globally one of the most common diseases from the middle age onwards. Cartilage is an avascular tissue therefore it cannot be repaired in the body. Conservative treatments have failed as a good remedy and cell therapy as a decisive cure is needed. One of the best and easily accessible cell sources for this purpose is adipose-derived stem cells which can be di...

Transcriptome analysis of early chondrogenesis in ATDC5 cells induced by bone morphogenetic protein 4.

We performed serial analysis of gene expression (SAGE) profiling in mouse chondrogenic ATDC5 cells before and 6 h after the onset of chondrogenesis induced by BMP4. A total of 43,656 SAGE tags (21,875 and 21,781 tags from the uninduced and induced libraries, respectively) were analyzed. Our analysis predicted that 139 transcripts were differentially represented in the two libraries (p < 0.05), ...
