چکیده مقاله:

Objective(s)Metformin is well known for activation of AMP-activated protein kinase (AMPK). AMPK activation inhibits mammalian target of rapamycin (mTOR) as a key signaling process in cell proliferation. Recent epidemiological studies demonstrate that metformin lowers the risk for several types of cancer in diabetic patients. Concerning the critical role of angiogenesis in the incidence and progression of tumors, we investigated the effect of metformin on human umbilical vein endothelial cells migration, as well as on vascular endothelial growth factor (VEGF) expressions in the cells and also on angiogenesis in air pouch model in rats.Materials and MethodsA "wound" repair method was used to assess the cell migration (n=6). Real-time PCR was performed to quantify the mRNA expression of VEGF (n=5). In air pouch model, carrageenan was injected into the air pouches on the back of rats (n=6) and following an IV injection of carmine red dye granulomatous tissue was processed for the assessment of the dye content. An ordinary ANOVA with Student-Newman-Keuls post hoc test was used to compare groups.ResultsMetformin (orally, 50mg/kg) significantly (P

Anti-Angiogenic Activity of Ficus carica Latex Extract on Human Umbilical Vein Endothelial Cells

Angiogenesis, the formation of new blood vessels, is a key process in cancer development and metastasis. In this study, the anti-angiogenic and anti-proliferative potentials of Ficus carica latex extract have been investigated using human umbilical vein endothelial cells (HUVECs). Different doses of latex extract were prepared and added to a three-dimensional culture of HUVEC in a collagen matr...

Effects of alpha-mangostin on memory senescence induced by high glucose in human umbilical vein endothelial cells

Objective(s): Hyperglycemia induces cellular senescence in various body cells, such as vascular endothelial cells. Since the vessels are highly distributed in the body and nourish all tissues, vascular damages cause diabetes complications such as kidney failure and visual impairment. Alpha-mangostin is a xanthone found in mangosteen fruit with protective effects in met...

Comparative Study of Anti-Cancer Properties of Hydroalcoholic Extract of Different Cultivars of Apricot Kernels on Breast Cancer Cells (MCF7) and Human Umbilical Vein Endothelial Cells

Background and purpose: Breast cancer mortality is growing around the world due to chemotherapy resistance and chemo-related side effects. Many natural products from plants are recognized to exert anticancer activity. This study aimed at investigating anti-proliferative and apoptotic activity of hydro-alcoholic extract of different varieties of apricot kernels (Jahangiri, Palmia, Jafari, and N5...

Anti-angiogenic action of plasma hyaluronan binding protein in human umbilical vein endothelial cells.

The kringle domain is a triple loop structure present in angiostatin and endostatin. The disulfide bond-linked kringle architectures have been known to be essential for anti-angiogenic activity. Plasma hyaluronan binding protein (PHBP) is a novel serine protease which consists of three epidermal growth factor (EGF) domains, a kringle domain, and a serine protease domain. PHBP can be cleaved aut...

Metabolomics of the effect of AMPK activation by AICAR on human umbilical vein endothelial cells.

AMP-activated protein kinase (AMPK) is a metabolic master switch expressed in a great number of cells and tissues. AMPK is thought to modulate the cellular response to different stresses that increase cellular AMP concentration. The adenosine analog, 5-aminoimidazole-4-carboxamide-1-β-D-ribofuranoside (AICAR) is an AMPK activator used in many stud...
