چکیده مقاله:

Valeriana officinalis (valerian), as a nutraceutical herb, is widely used for its sedative and hypnotic properties. It is known that C15 sesquiterpenoid valerenic acid (VA) is the active ingredient responsible for pharmacological effects of V. officinalis. To evaluate the effect of methyl jasmonate (MeJA) concentrations (50 and 100 µM) in the modulation of expression patterns of the genes involved in valerenic acid (VA) biosynthesis, transcript abundance of identified sesquiterpene synthase (Sesqui-TPS) genes in the root of V. officinalis was monitored by quantitative real time PCR (qRT-PCR) within a 144 h time period. In addition, Valerenic acid contents were measured by high-performance liquid chromatography (HPLC). The highest amount of VA (12.45 mg/g dry weight (DW)) was found at 100 µM MeJA with a 12 fold increase over control culture (1.03 mg/g DW) at exposure time of 72 h. Moreover, MeJA in a concentration dependent manner enhanced transcription rate of VoTPS1 and VoTPS7 genes. Accordingly, exposure to 100 µM MeJA for 24 h can be more effective in the induction of these genes than that observed for 50 µM. Such enhancement was correlated with increased VA accumulation suggesting that these genes may be responsible for the biosynthesis of intermediates involved in the VA-biosynthetic pathway. However, MeJA treatment seemed to have a less significant effect on VoTPS3 expression than VoTPS1 and VoTPS7 genes. This results provide insights for more effective biosynthesis of VA by MeJA-mediated transcriptional changes of putative sesqui-TPS.

Enhanced production of valerenic acids and valepotriates by in vitro cultures of Valeriana officinalis L.

Valerian (Valeriana officinalis L.) is an important medicinal plant that is widely exploited for its roots and rhizomes which contain valepotriates and valerenic acids (with putative pharmacological activities). Thus root proliferation of valerian is very important. The aim of this study was to establish a practical tissue culture method for rapid and large-scale induction of V. officinalis L. ...

Metabolic engineering of Escherichia coli for production of valerenadiene.

Valeriana officinalis is a medicinal herb which produces a suite of compounds in its root tissue useful for treatment of anxiety and insomnia. The sesquiterpene components of the root extract, valerenic acid and valerena-1,10-diene, are thought to contribute to most of the observed anxiolytic of Valerian root preparations. However, valerenic acid and its biosynthetic intermediates are only prod...

Production of sesquiterpene-type phytoalexins by hairy roots of Hyoscyamus albus co-treated with cupper sulfate and methyl jasmonate.

The production of sesquiterpene-type phytoalexins with a vetispyradiene skeleton by Hyoscyamus albus hairy roots induced by methyl jasmonate (MeJA) was reported in a previous paper. The production pattern on co-treatment with cupper sulfate and MeJA (CuSO(4)-MeJA) showed a TLC profile differing from that on treatment with MeJA. Thus, we studied the production of phytoalexins on hairy root cultu...

Valerenic acid and Valeriana officinalis extracts delay onset of Pentylenetetrazole (PTZ)-Induced seizures in adult Danio rerio (Zebrafish)

BACKGROUND Anticonvulsant properties have been attributed to extracts of the herbal medicine Valeriana officinalis. Our aims were to examine the anticonvulsant properties of valerenic acid and valerian extracts and to determine whether valerian preparations interact with the activity of other anti-epileptic drugs (phenytoin or clonazepam). To achieve these goals, we validated the adult zebrafis...

Selective Interactions of Valeriana officinalis Extracts and Valerenic Acid with [3H]Glutamate Binding to Rat Synaptic Membranes

Although GABA neurotransmission has been suggested as a mechanism for Valeriana officinalis effects, CNS depression can also be evoked by inhibition of ionotropic (iGluR) and metabotropic glutamate receptors (mGluR). In this study, we examined if aqueous valerian extract interacted with glutamatergic receptors. Freshly prepared aqueous valerian extract was incubated with rat cortical synaptic m...
