چکیده مقاله:

ABSTRACT             Background and objectives: Pterygium is a non-cancerous growth of conjunctival tissue that can extend onto the corneal surface. The presence of some oncogenic viruses in pterygium and the neoplastic nature of these lesions led us to the postulated involvement of the viruses in the etiology of pterygium. Given the association of human herpesvirus 6 (HHV-6) with ocular diseases, we aimed to investigate presence of this virus in pterygium.             Methods: Fifty tissue specimens were collected from patients with pterygium who underwent pterygium surgery between February 2013 and May 2015. The specimens were tested by real-time PCR using Maxima SYBR Green/ROX qPCR Master Mix (2X) kit. Demographic and clinical data were collected and analyzed using SPSS software (version 18).             Results: Six (12 %) specimens were positive for HHV-6 DNA. There was no statistically significant correlation between pterygium and presence of HHV-6.             Conclusion: Based on the results, a direct association between HHV-6 and development of pterygium seems less probable, which suggests that other etiologic agents must be involved in the multistep process of the disease.             Keywords: Human Herpesvirus 6; pterygium; Real-time PCR.

Optimization of the Sybr Green real time PCR for the detection of Human Herpes Virus type 6 (HHV-6).

HHV-6 is the etiological agent of Exanthem subitum which is considered the sixth most frequent disease in infancy. In immuno-compromised hosts, reactivation of latent HHV-6 infection may cause severe acute disease. We developed a Sybr Green Real Time PCR for HHV-6 and compared the results with nested conventional PCR. A 214 pb PCR derived fragment was cloned using pGEM-T easy from Promega syste...

Comparison of the SYBR Green and the hybridization probe format for real-time PCR detection of HHV-6.

A comparative study was conducted of a novel real-time quantitative PCR test (LightCycler System) with FastStart DNA Master(PLUS) SYBR Green I dye to detect DNA of human herpes virus 6 (HHV-6). Results were compared with those of a real-time quantitative PCR with hybridization probe (HP) formats using the fluorescence resonance energy transfer method, and with those of a single qualitative PCR ...

Evaluation of Epstein-Barr virus, human herpesvirus 6 (HHV-6), and HHV-8 antiviral drug susceptibilities by use of real-time-PCR-based assays.

Evaluation of candidate antiviral drugs against Epstein-Barr virus (EBV), human herpesvirus 6 (HHV-6), and HHV-8 is hampered by the lack of convenient laboratory assays. We developed real-time quantitative PCR assays performed on supernatants of lymphoma cell lines and determined the 50% inhibitory concentrations (IC50s) of nucleoside, nucleotide, and pyrophosphate analogues against these herpe...

Development of SYBR Green I Based Real-Time RT-PCR Assay for Specific Detection of Watermelon silver mottle Virus

Background: Watermelon silver mottle virus (WSMoV), which belongs to the genus Tospovirus, causes significant loss in Cucurbitaceae plants. Objectives: Development of a highly sensitive and reliable detection method for WSMoV. Materials and Methods: Recombinant plasmids for targeting the sequence of nucleocapsid protein gene of WSMoV were constructed. SYBR Green I real-time PCR was established...

Real-time PCR for quantitation of bovine viral diarrhea virus RNA using SYBR Green I fluorimetry

Quantitative real-time RT-PCR (qRT-PCR) assay was developed for the detection and quantification of bovine viral diarrhea virus (BVDV) in clinical samples from persistently infected cattle. qRT-PCR was optimized to quantify the number of BVD virus copies using Light Cycler® detection system and intercalation fluorogenic dye SYBR Green I. A universal set of primers was selected from a highly con...

The effect of human herpesvirus-6 (HHV-6) on cultured human neural cells: oligodendrocytes and microglia.

Human herpesvirus-6 (HHV-6) is a betaherpesvirus that has been frequently associated with pediatric encephalitis. In 1995 Challoner et al reported that HHV-6 variant B (HHV-6B) was linked to multiple sclerosis (MS) due to the presence of viral DNA and antigen in the oligodendrocytes surrounding MS plaques. These findings led us to examine HHV-6B's in vitro tropism for primary neural cells. HIV-...
