چکیده مقاله:

Introduction: Mesenchymal stem cells (MSCs) are suitable candidates for the treatment of liver diseases. However, their low survival rate limits their efficacy following transplantation. This study aimed to evaluate the therapeutic potentials of H2O2-preconditioned umbilical cord-derived MSCs (UCMSCs) on acute liver failure (ALF) in mice. Methods: UCMSCs were pre-conditioned with different concentrations of H2O2. Cell viability was evaluated by WST-1 (water soluble tetrazolium) assay followed by exposure to lethal doses of H2O2. ALF was induced in NMRI mice using CCl4 and the cells therapy was performed using H2O2-preconditioned and normal UCMSCs. After 24, 48, and 72 hours, regenerative potentials of different UCMSCs groups were evaluated compared to sham group (that receive no MSCs) using biochemical and histological methods.  Results: Lower liver enzymes was significantly evident in mice transplanted with H2O2-preconditioned UCMSCs compared with the other groups. Interestingly, histological results revealed a significant improvement in liver regeneration in these mice. Conclusion: Preconditioning of UCMSCs with H2O2 not only enhances their survival but also increases the efficacy of MSCs-based cell therapy in acute liver failure.

Autophagy-Modulated Human Bone Marrow-Derived Mesenchymal Stem Cells Accelerate Liver Restoration in Mouse Models of Acute Liver Failure

Background: Mesenchymal stem cells (MSCs) have been recently received increasing attention for cell-based therapy, especially in regenerative medicine. However, the low survival rate of these cells restricts their therapeutic applications. It is hypothesized that autophagy might play an important role in cellular homeostasis and survival. This study aims to investigate the regenerative potentia...

Transplantation of umbilical cord mesenchymal stem cells via different routes in rats with acute liver failure.

OBJECTIVE This study aimed to compare the therapeutic efficacy of transplantation of human umbilical cord mesenchymal stem cells (hUCMSC) in different routes in acute hepatic failure (ALF) in rats. METHODS hUCMSCs were isolated and identified by detection of surface antigens via flow cytometry. In T group and H group, ALF rats received hUCMSC transplantation through the tail vein and intrahep...

Tonsil-derived Mesenchymal Stem Cells Ameliorate CCl4–induced Liver Fibrosis in Mice via Autophagy Activation

Liver transplantation is the treatment of choice for chronic liver failure, although it is complicated by donor shortage, surgery-related complications, and immunological rejection. Cell transplantation is an alternative, minimally invasive treatment option with potentially fewer complications. We used human palatine tonsil as a novel source of mesenchymal stem cells (T-MSCs) and examined their...

Human umbilical cord blood-derived mesenchymal stem cells ameliorate psoriasis-like skin inflammation in mice

Mesenchymal stem cells (MSCs) inhibit the proliferation or activation of lymphocytes, and their inhibitory effects do not require human leukocyte antigen (HLA)-matching because MSCs express low levels of HLA molecules. Therefore, MSCs may be able to regulate immune responses. In this study, we determined whether MSCs could inhibit psoriasis-like skin inflammation in mice. After induction of pso...

Therapeutic Potential of HGF-Expressing Human Umbilical Cord Mesenchymal Stem Cells in Mice with Acute Liver Failure.

Human umbilical cord-derived mesenchymal stem cells (UCMSCs) are particularly attractive cells for cellular and gene therapy in acute liver failure (ALF). However, the efficacy of this cell therapy in animal studies needs to be significantly improved before it can be translated into clinics. In this study, we investigated the therapeutic potential of UCMSCs that overexpress hepatocyte growth fa...

Regenerative Effect of Bone Marrow-derived Mesenchymal Stem Cells in Thioacetamide-induced Liver Fibrosis of Rats

The present study determined the regenerative effect of bone marrow-derived stem cells (BMSCs) on thioacetamide (TA)-induced liver fibrosis in rats. A total of 30 male Wistar rats were randomly divided into sham control and treatment groups. The rats of the sham control group were subdivided into three groups and sampled on the 14th, 18th, and 20th weeks after fibrosis induction. The rats of th...
