چکیده مقاله:

A protocol was developed for high frequency in vitro multiplication of an ornamental orchid, Phalaenopsis amabilis (L.) Blume var. Jawa, using plant growth regulators (PGRs). Protocorm-like bodies (PLBs), as explants were cultured on Murashige and Skoog (MS) medium fortified with various concentrations of kinetin (KIN) and indole-3-butyric acid (IBA), either individually or in combination. A combination of 1.00 mg l-1 KIN + 1.00 mg l-1 IBA was found to be suitable for regeneration of most measured characteristics especially maximum PLBs regeneration (30.40/plantlet), leaf number (5.93/plantlet), and root number (8.36/plantlet) from protocorm explants. The maximum number of plantlets (11.66) was calculated on MS medium supplemented with 1.00 mg l-1 KIN + 0.50 mg l-1 IBA, followed by 1.00 mg l-1 KIN + 1.00 mg l-1 IBA (10.33). Plantlets were transplanted to pots filled with cocochips and sphagnum moss (70:30) for acclimatization and transferred to the greenhouse. Upon ex vitro transfer, 90% of plants survived.

Clonal Propagation of Phalaenopsis amabilis (L.) BL. cv. 'Golden Horizon' Through In vitro Culture of Leaf Segments

A protocol was established for mass clonal propagation of Phalaenopsis amabilis cv. 'Golden horizon' through in vitro culture of young leaf segments from mature plant. Explants were cultured on half strength Murashige and Skoog (1/2 MS) medium supplemented with N6-benzyladenine (2.0 mg l-1), a-naphthaleneaceetic acid (0.5 mg l-1), 2% (w/v) sucrose, 10% (v/v) coconut water, 2 g l-1 peptone and 1...

Phenolic Compounds and Antioxidant Activity of Phalaenopsis Orchid Hybrids

Phalaenopsis spp. is the most commercially and economically important orchid, but their plant parts are often left unused, which has caused environmental problems. To date, reports on phytochemical analyses were most available on endangered and medicinal orchids. The present study was conducted to determine the total phenolics, total flavonoids, and antioxidant activity of ethanol extracts prep...

Interactions of B-class complex proteins involved in tepal development in Phalaenopsis orchid.

In our previous studies, we identified four DEFICIENS (DEF)-like genes and one GLOBOSA (GLO)-like gene involved in floral organ development in Phalaenopsis equestris. Revealing the DNA binding properties and protein-protein interactions of these floral homeotic MADS-box protein complexes (PeMADS) in orchids is crucial for the elucidation of the unique orchid floral morphogenesis. In this study,...

In Vitro Propagation of Lisianthus (Eustomagr andiflurom)

Nowadays, the most rapid method for producing healthy and disease-free Lisiantus is micropropagation. With respect to high economic value of this plant which is regarded among the 10 top cutting flowers in the world, this research was carried out to suggest a suitable protocol for its in vitro propagation, using nodal sections as an explant. To carry out this object, the effects of the pH (5.5,...

Optimizing in vitro propagation of Rosa persica

Rosa persica with a distinctive red eye is a good candidate for addition to the breeding pool of roses. In order to optimize micropropagation of R. persica, three culture media (MS, QL and VS) were compared. The maximum growth rate was observed in the VS medium. Addition of FeEDTA (Fe: 65.4 mg/l), calcium gluconate (Cg: 2.7 g/l) and Gamborg vitamins (B5) instead of VS vitamins, alone or in comb...
