چکیده مقاله:

Background: Preeclampsia is one of the most common complications of pregnancy that occurs after the 20th weeks of pregnancy. The pathophysiology of this disease is not exactly known. Transforming Growth Factor-beta (TGF-β) and Nitric Oxide (NO) are thekey regulatory factors secreted by Mesenchymal stem Cells (MSCs). Objectives: The aim of the present study was to evaluate the TGF-β and NO secretion by adipose-derived MSCs in normal andpreeclamptic pregnant women. Materials and methods: The adipose tissues were collected from 10 preeclamptic patients and 10 age-matched normotensivecontrols at the time of cesarean section delivery. After isolation and expansion of MSCs, their capability of differentiation andimmunophenotyping characteristics were assessed. Next, the release of TGF-ß was evaluated making use of ELISA sandwichmethod and Griess method was used to measure the level of NO. Results: Adipose derived MSCs in both groups were differentiated into osteocytes and adipocytes. The expression of CD90, CD73,CD44, and CD105 markers and lack of expression of CD-14, CD34, CD45, and HLA-DR markers in cells isolated from adipose in bothgroups was observed using flow cytometric analysis. The levels of TGF-ß secretion in preeclamptic women were significantly higherthan those in control group, but the mean level of NO secreted by adipose derived MSCs did not significantly change in the twogroups. Conclusion: It can be concluded that significant increase in the secretion of TGF-β owing to MSCs in preeclamptic participantsshows the importance of these cells in controlling immunological balance in these patients. Therefore, MSCs-based therapy seemsto regulate TH1/Th2 balance in preeclampsia.

Comparison of Characteristics of Human Amniotic Membrane and Human Adipose Tissue Derived Mesenchymal Stem Cells

BACKGROUND Mesenchymal stem cells (MSCs) are ideal candidates for treatment of diseases. Amniotic membranes are an inexpensive source of MSCs (AM-MSC) without any donor site morbidity in cell therapy. Adipose tissue derived stem cells (ASCs) are also suitable cells for cell therapy. There is discrepancy in CD271 expression among MSCs from different sources. In this study, the characteristics...

Comparative analysis of the Gene expression profile of Chemokine Receptors between Adipose-derived and Bone marrow-derived Mesenchymal Stem Cells

Introduction: Mesenchymal stem cells (MSCs) hold great promise in the field of regenerative medicine.Although originally isolated from bone marrow, MSCs have since been obtained from a variety of adult and neonatal tissues including the adipose tissue. Stemness and multipotential features of Mesenchymal Stem Cells (MSC) has been highlighted in many studies but there are many dark aspects in ex...

Nitric Oxide Production and Apoptosis Induction in T Cells through Stimulation of Mesenchymal Stem Cells with Zymosan

Background & Aims: Mesenchymal stem cells (MSCs) are non-hematopoietic multipotant cells, which multiply through attaching to culture plates. Toll-like receptors (TLRs) are inherent immune sensors and regulators of immunomodulatory activities of MSCs. The aim of this study was to investigate the effects of zymosan on stem cell polarization into anti-inflammatory phenotypes through the productio...

Gene manipulation of human adipose-derived mesenchymal stem cells by miR-34a

Background: Safe and effective gene therapy is considered as one of the therapeutic goals in many diseases. Due to the important role of stem cells in cell therapy, this study aimed to produce human adipose-derived mesenchymal stem cells (hASCs) using the miR-34a overexpression. Materials and methods: The hsa-mir-34a precursor sequence was cloned into the PCDH lentiviral vector. The recombinant...
