I-34: Interactorme of Human Embryo Implan Implantation:Pathways,Networks

نویسنده

چکیده مقاله:

Background: A prerequisite for successful embryo implantation is adequate preparation of receptive endometrium and the establishment and maintenance of a viable embryo. The success of implantation further relies upon a two-way dialogue between the embryo and uterus. However, molecular bases of these preimplantation and implantation processes in humans are not well known. Materials and Methods: We performed genome expression analyses of human embryos (n = 128) and human endometria (n = 8). We integrated these data with proteinprotein interactions in order to identify molecular networks within the endometrium and the embryo, and potential embryo-endometrium interactions at the time of implantation. For that we applied a novel network profiling algorithm HyperModules, which combines topological module identification and functional enrichment analysis. Results: We found a major wave of transcriptional downregulation in preimplantation embryos. In receptive-stage endometrium, several genes and signalling pathways were identified, including JAK-STAT signalling and inflammatory pathways. The main curated embryo-endometrium interaction network highlighted the importance of cell adhesion molecules in the implantation process. We also identified cytokine-cytokine receptor interactions involved in implantation, where osteopontin, LIF and LEP pathways were intertwining. Further, we identified a number of novel players in human embryo-endometrium interactions, such as APOD, EDN1, FGF7, GAST, KREMEN1, NRP1, SERPINA3, VCAN, and others. Conclusion: Our findings provide a fundamental resource for better understanding of the genetic network that leads to successful embryo implantation. We demonstrate the first systems biology approach into the complex molecular network of the implantation process in humans. I-26: Considerations for PGD Applications inElective Human Embryo Sex Selection

The promise of medical innovation has long evoked social commentary, particularly when personal reproductiveautonomy may be impacted. Development of the oral contraceptive, effective and safe surgical sterilization, and later IVF and ICSI are among the revolutionary developments where initial reaction was dubious, but eventually, each was accorded mainstream status as clinical experience accumu...

I-21: Embryo Relinquishment for Reproduction

Background: The conceptualization of the transfer of embryos between the individuals who created them to one or more recipients for family-building is hotly contested – particularly as regards whether the practice should be most appropriately considered to be “donation” or “adoption”. This paper examines this debate, considering the research carried out on the intentions and decisions of those ...

I-5: Cancellation of Fresh Embryo Transfer: A Future Perspective

Controlled ovarian hyperstimulation (COH) affects adversely implantation following in vitro fertilization and embryo transfer. COH with gonadotropins alters endometrial receptivity and luteal function. In addition periovulatory endometrial characteristics in stimulated cycles are considerably different compared with the natural cycles, and periovulatory secretory transformation is consistently ...

I-9: Embryo Assessment and Selection

Over the years it has been an important goal to optimize the treatment of subfertility in order to obtain a better pregnancy rate and baby-take-home rate, usually combined with the goal of decreasing the rate of multiplet pregnancies. This implies optimizing the development of embryos through improved in vitro culture methods. And it also implies optimizing the methods of selection of embryos f...

I-12: Objective Embryo Assessment Utility of Time-Lapse

Background Traditionally, embryo incubation and assessment daily has been under a light microscope, these observations are inevitably restricted to specific times and considering that the development of the embryo is a dynamic process, several critical stages in between observations may go unnoticed. For this reason, the new technologies, time lapse monitoring, have focused on the research for ...
