Merozoite surface proteins (MSPs) of malaria parasites play critical roles during the erythrocyte invasion and so are potential candidates for malaria vaccine development. However, because MSPs are often under strong immune selection, they can exhibit extensive genetic diversity. The gene encoding the merozoite surface protein-3 (MSP-3) of Plasmodium falciparum displays 2 allelic types, K1 and ...
Severe Malaria is a medical emergency mainly because of its rapid progression to complications and death if not promptly adequately treated. In 2018 WHO put the incidence P. falciparum in African region be around 99.7%.
A mevalonate-independent pathway of isoprenoid biosynthesis present in Plasmodium falciparum was shown to represent an effective target for chemotherapy of malaria. This pathway includes 1-deoxy-D-xylulose 5-phosphate (DOXP) as a key metabolite. The presence of two genes encoding the enzymes DOXP synthase and DOXP reductoisomerase suggests that isoprenoid biosynthesis in P. falciparum depends o...
Evaluation of a colorimetric PCR-based assay to diagnose Plasmodium falciparum malaria in travelers.
New diagnostic tests are needed to facilitate the diagnosis of Plasmodium falciparum malaria in the returned traveler. We performed a blinded evaluation of a nonisotopic colorimetric PCR-based assay (Digene SHARP Signal System) and compared the results with those obtained by microscopy and nested PCR for the detection of P. falciparum malaria in 150 febrile travelers. By using nested PCR as the...
The combination of anemia in malarial infestations ranges from nutritional deficiency to marrow suppression. An immune hemolytic anemia in Plasmodium falciparum malaria is also a part of this wide-ranging spectrum. But the co-existence of cold agglutinins in a Plasmodium falciparum malaria has only sporadically been reported in literature. We present a case of a patient with falciparum malaria ...
The malaria parasite, Plasmodium falciparum, exploits multiple ligand-receptor interactions, called invasion pathways, to invade the host erythrocyte. Strains of P. falciparum vary in their dependency on sialated red cell receptors for invasion. We show that switching from sialic acid-dependent to -independent invasion is reversible and depends on parasite ligand use. Expression of P. falciparu...
BACKGROUND Areas endemic for Plasmodium falciparum, hepatitis B virus (HBV) and hepatitis C virus (HCV) overlap in many parts of sub-Saharan Africa. HBV and HCV infections develop in the liver, where takes place the first development stage of P. falciparum before its further spread in blood. The complex mechanisms involved in the development of hepatitis may potentially influence the developmen...
OBJECTIVES Studies have typically investigated health and educational consequences of malaria among school-aged children in areas of high malaria transmission, but few have investigated these issues in moderate transmission settings. This study investigates the patterns of and risks for Plasmodium falciparum and anaemia and their association with cognitive and education outcomes on the Kenyan c...
Intercellular adhesion molecule 1 (ICAM-1) is an endothelial cell adhesion molecule implicated in cerebral malaria. We investigated whether fibrinogen affects Plasmodium falciparum binding to ICAM-1, as the ICAM-1 binding sites of P. falciparum and fibrinogen overlap. We show that fibrinogen dramatically reduces P. falciparum adhesion to ICAM-1 under flow conditions.
