RT-LAMP法を用いて,ナシ(Pyrus spp.)の葉からナシ葉退緑斑点随伴ウイルス(PCLSaV)を検出する技術を開発した。設計したプライマーセットのうち,R2LP4が最も増幅が早く,退緑斑点症状のあるナシ葉の全RNAから15分以内にPCLSaVが検出された。12県で採取されたニホンナシおよびセイヨウナシの葉を用いた診断では,RT-LAMP法は既報のRT-PCR法と同等の感度と精度を示した。さらに,注射針で採取した汁液を用いた簡易なRT-LAMP検定においてもPCLSaVを検出することができた。また,塩基配列情報の比較から,設計したプライマーの一部は,日本以外で発生が確認されているPCLSaVの中国分離株と適合する可能性が示された。
