1. A Sepharose-(glutathione-2-pyridyl disulphide) conjugate has been prepared. 2. Its use in a new type of chromatography, covalent chromatography by thiol-disulphide interchange, is described. 3. With this technique, papain containing 1 intact catalytic site [thiol with high reactivity towards 2,2'-dipyridyl disulphide (2-Py-S-S-2-Py) at pH4] per mol of protein is readily prepared both from dr...

XCP1 is a xylem-specific papain-like cysteine peptidase in Arabidopsis. To determine whether XCP1 could be involved in tracheary element autolysis, promoter activity and localization of XCP1 were investigated using XCP1 promoter-beta-glucuronidase fusions and immunofluorescence confocal microscopy. A tracheary element expression pattern was detected for XCP1. Results from confocal microscopy an...

A microvillar fraction was prepared from human kidney cortex. This fraction was seven to 10 times enriched in aminopeptidases N and A, gamma-glutamyltransferase, dipeptidyl peptidase IV, neutral endopeptidase and alkaline phosphatase. Dipeptidyl peptidase IV activity of human renal microvilli could be inhibited by di-isopropylphosphorofluoridate and neutral endopeptidase activity by phosphorami...

BACKGROUND New approaches for chemomechanical caries removal require effective materials with antibacterial properties for removal of infected dentin. Apacaries gel is a newly developed material comprised polyphenol from mangosteen extracts and papain mixed in gel preparation. AIM This study evaluated the antibacterial effects of Apacaries gel on Streptococcus mutans in vitro. MATERIALS AND...

Azo-dye was liberated from azocoll not only by proteolytic enzymes but also by albumin (serum or egg), a culture medium (papain digest of meat) or polyvinylpyrrolidone (PVP). Pretreatment of azocoll with papain and PVP increased the reactivity of azocoll to ƒÈ-toxin of Clostridium perfringens. A rapid and reliable assay method of ƒÈ-toxin and ƒÈ-antitoxin using the pretreated azocoll is described.

Pathogenic bacteria deliver effector proteins into host cells through the type III secretion apparatus to modulate the host function. We identify a family of proteins, homologous to the type III effector Cif from enteropathogenic Escherichia coli, in pathogens including Yersinia, Photorhabdus, and Burkholderia that contain functional type III secretion systems. Like Cif, this family of proteins...

Evidence has been presented earlier (2, 3) that leucine aminopeptidase can degrade about two-thirds of the molecule of mercuripapain, liberating free amino acids, without altering the enzymic activity of the papain when it is reactivated. Although several experimental results supported this conclusion, the best evidence came from two types of observations: first, new amino end groups different ...

We have examined the secondary structures of human class I and class II histocompatibility antigens in solution by Fourier transform infrared spectroscopy and circular dichroism in order to compare the relative amounts of alpha-helix, beta-sheet, and other structures, which are crucial elements in the comparison of the protein structures. Quantitation of infrared spectra of papain-solubilized H...

Papain-like protease is a SARSCOV-2 that functions for ubiquitination in host cells. Caffeine compounds have been widely reported to antioxidant, anti-inflammatory, antidiabetic, and antiobesity activities. However, hexose malic acid coffee rind as potential inhibitors of the papain-like SARSCOV2 not reported. This study investigated PLPro inhibitor agents inhibiting ubiquitination. In silico s...

Investigations are in progress in this laboratory to elucidate the amino acid sequence of the proteolytic enzyme, papain (2). Results have already been presented on the isolation and sequence of some peptides obtained after chymotryptic digestion of oxidized papain (3-5). In the present paper, we report the isolation, composition, and sequence of some peptides produced by tryptic digestion of o...
