OBJECTIVE Posaconazole prophylaxis during induction chemotherapy for acute myeloid leukaemia (AML) and myelodysplastic syndromes (MDS) has been shown to significantly decrease the incidence of invasive fungal disease (IFD) and increase overall survival in a trial setting, but only small real-life studies have been published. METHODS This was a retrospective cohort study including consecutive ...
background candiduria is a rising condition among hospitalized patients and candida albicans is the most common recovered agent. however, non-albicans candida species (nacs) such as c. glabrata, c. krusei, c. parapsilosis, and c. tropicalis are also important. although most candida species especially c. albicans are sensitive to routinely used antifungals, an increasing trend in resistance has ...
The aim of this single-center, phase 1, randomized, 5 by 5 crossover, open-label study was to determine the effects of varying amounts of a nutritional supplement (Boost Plus) on the pharmacokinetics of posaconazole in 30 healthy volunteers. After an overnight fast, subjects were administered a single dose of 400 mg posaconazole oral suspension alone or following Boost Plus (8 fluid ounces [oz]...
We thank Dr. Blennow and Dr. Kalin for their comments (1) relating to our recent paper (2). They argue that some of the cases included in our study as breakthrough fungal infections occurring while patients were taking posaconazole prophylaxis are not cases of “invasive” fungal infections (IFIs). To clarify, we agree that case 4 (culture-proven oral candidiasis), case 7 (culture-proven urinary ...
Posaconazole (PCZ) is frequently used for prophylaxis against invasive fungal infections (IFI) in patients undergoing induction chemotherapy for acute myeloid leukemia (AML) or myelodysplastic syndrome (MDS). Posaconazole is commercially available as an oral suspension (PCZ-susp) and as a delayed-release tablet (PCZ-tab). Differences in absorption and bioavailability between these formulations ...
The posaconazole MIC(90) for 1,903 yeast isolates from France was 1 microg/ml (range, < or =0.015 to 8 microg/ml). Ninety percent of isolates with fluconazole MICs of > or =8 microg/ml (n = 509) and 90% of those with voriconazole MICs of > or =2 microg/ml (n = 80) were inhibited by 2 and 8 microg/ml of posaconazole, respectively.
Aspergillus terreus is a species which is being seen increasingly frequently and which is highly resistant to amphotericin B in vitro and clinically. We evaluated amphotericin B, caspofungin, and posaconazole in a murine model of acute invasive aspergillosis. Caspofungin and posaconazole both appeared beneficial and may be reasonable treatment alternatives for infection with A. terreus.
The activities of 10 combinations of antifungal agents against 25 clinical isolates of Scopulariopsis brevicaulis were tested by the checkerboard technique. An average indifferent effect was detected for all combinations. Synergy was observed for some isolates and combinations, particularly with posaconazole-terbinafine (68% of strains), amphotericin B-caspofungin (60%), and posaconazole-caspof...
The in vitro activities of amphotericin B, itraconazole, fluconazole, voriconazole, posaconazole, and ravuconazole against 39 isolates of Trichosporon spp. were determined by the NCCLS M27-A microdilution method. The azoles tested appeared to be more potent than amphotericin B. Low minimal fungicidal concentration/MIC ratios were observed for voriconazole, posaconazole, and ravuconazole, sugges...
BACKGROUND We evaluated the cost-effectiveness of posaconazole compared with standard azole therapy (SAT; fluconazole or itraconazole) for the prevention of invasive fungal infections (IFI) and the reduction of overall mortality in high-risk neutropenic patients with acute myelogenous leukaemia (AML) or myelodysplastic syndromes (MDS). The perspective was that of the Spanish National Health Ser...
