Quantification of membrane partition potential of drug compounds is of great pharmaceutical interest. Here, a novel approach combining liquid-state NMR diffusion measurements and fast-tumbling lipid/detergent bicelles is used to measure accurately the partition coefficient K(p) of amantadine in phospholipid bilayers. Amantadine is found to have a strong membrane partition potential, with K(p) o...

We report the case of a 69-year-old woman with seizures and acute renal failure with hyperkalemia. She presented with bladder turgescence and hydronephrosis on admission and was diagnosed as obstructive acute renal failure. Urethral catheterization was performed after a single-session hemodialysis. It resulted in immediate improvement of renal function and consciousness, and subsequent disappea...

N-methyl-D-aspartate (NMDA) receptor blockade can improve L-DOPA (l-3,4-dihydroxyphenylalanine)-induced dyskinesias in Parkinson’s disease (PD) patients. Amantadine is a well-tolerated and effective antiparkinsonian agent, recently found to possess NMDA antagonistic properties. Oxidative damage may contribute dopaminergic (DAergic) neurodegeneration the substantia nigra of patients with PD. N,N...

Influenza A virus infects many species, and amantadine is used as an antiviral agent. Recently, a substantial increase in amantadine-resistant strains has been reported, most of which have a substitution at amino acid position 31 in the M2 gene. Understanding the mechanism responsible for the emergence and spread of antiviral resistance is important for developing a treatment protocol for seaso...

OBJECTIVE To evaluate the efficacy of amantadine in the treatment of symptoms and neurocognitive performance in adolescents following sports-related concussion. PARTICIPANTS A clinical sample of 25 male (n = 11) and female (n = 14) adolescent subjects with an age-, sex-, and concussion history-matched group of 25 male (n = 11) and female (n = 14) control subjects. SETTING Outpatient concuss...

Background: Recent short-term studies suggested that amantadine (Ama) might ameliorate dyskinesia in patients with Parkinson’s disease. A double-blind study programmed over 12 months was designed to assess the duration of the antidyskinetic effect of amantadine on levodopa induced dyskinesia. Methods: 40 patients treated for 7.5 (2.2) years with levodopa (729.3 (199.4) mg/day) and dopaminoagoni...

The role of dopamine as a possible central inhibitory mediator of pain processes has been demonstrated. The administration of L-Dopa diminishes pain perception in humans as well as the response to nociceptive stimuli in animals. Also, the intracerebral microinjection of dopamine in inflammatory and neuropathic pain models (formalin test and deafferentation, respectively) reduces nociceptive res...

Influenza virus H9N2 subtype has triggered co-infection with other infectious agents, resulting in huge economical losses in the poultry industry. Our current study aims to evaluate the antiviral activity of protocatechuic acid (PCA) against a virulent H9N2 strain in a mouse model. 120 BALB/c mice were divided into one control group, one untreated group, one 50 mg/kg amantadine hydrochloride-tr...

Surveillance of amantadine and oseltamivir resistance among influenza viruses was begun in Hong Kong in 2006. In 2008, while both A/Brisbane/59/2007-like and A/Hong Kong/2652/2006-like viruses (H1N1) were cocirculating, we detected amantadine and oseltamivir resistance among A/Hong Kong/2652/2006-like viruses (H1N1), caused by genetic reassortment or spontaneous mutation.

BACKGROUND Prior to 2007, highly pathogenic avian influenza (HPAI) H5N1 viruses isolated from poultry and humans in Vietnam were consistently reported to be clade 1 viruses, susceptible to oseltamivir but resistant to amantadine. Here we describe the re-emergence of human HPAI H5N1 virus infections in Vietnam in 2007 and the characteristics of the isolated viruses. METHODS AND FINDINGS Respir...
