We have developed a method for assembling genetic pathways for expression in Saccharomyces cerevisiae. Our pathway assembly method, called VEGAS (Versatile genetic assembly system), exploits the native capacity of S. cerevisiae to perform homologous recombination and efficiently join sequences with terminal homology. In the VEGAS workflow, terminal homology between adjacent pathway genes and th...

Nucleo-mitochondrial interactions, particularly those determining the primary divergence of biological species, can be studied by means of xenomitochondrial cybrids, which are cells where the original mitochondria are substituted by their counterparts from related species. Saccharomyces cerevisiae cybrids are prepared simply by the mating of the ρ(0) strain with impaired karyogamy and germinati...

Nowadays, the increasing interest in new market demand for alcoholic beverages has stimulated research on useful strategies to reduce ethanol content beer. In this context, use of non-Saccharomyces yeasts produce low-alcohol or alcohol-free beer may provide an innovative approach market. our study, four wild yeasts, belonging Torulaspora delbrueckii, Candida zemplinina and Zygosaccharomyces bai...

Rice wine is an alcoholic beverage of cereal produced as a result fermentation rice by selected yeasts isolated from different substrates. In this study the effects starter cultures on physico-chemical and sensory properties were investigated. The belong to six genera these are Saccharomyces, Kluveromyces, Schizosaccharomyces, Candida, Rhodotorula Geotrichum. species included S. cerevisi...

A three-plasmid yeast expression system utilizing the portable small ubiquitin-like modifier (SUMO) vector set combined with the efficient endogenous yeast protease Ulp1 was developed for production of large amounts of soluble functional protein in Saccharomyces cerevisiae. Each vector has a different selectable marker (URA, TRP, or LEU), and the system provides high expression levels of three ...

The following three elements were previously shown to be required for 3'-end formation of mRNA in the yeast Saccharomyces cerevisiae: (i) the efficiency element TATATA or related sequences, which function by enhancing the efficiency of downstream positioning elements; (ii) the positioning element AATAAA or related sequences, which position the poly(A) site; and (iii) the actual poly(A) site, wh...

We discovered two percolation processes in succession in dc conductivity of bulk baker's yeast in the course of dehydration. Critical exponents characteristic for the three-dimensional network for heavily hydrated system, and two dimensions in the light hydration limit, evidenced a dramatic change of the water network dimensionality in the dehydration process.
