A catalyst, solvent, work-up and column free synthesis of chromenopyranpyrazoles via multicomponent cascade reaction has been achieved with high stereoselectivity. This novel reaction creates two N-C, two C-C and one O-C bonds through a domino process for the construction of three new rings and three contiguous stereogenic centers.
The first total synthesis of (-)-maoecrystal Z is described. The key steps of the synthesis include a diastereoselective Ti(III)-mediated reductive epoxide coupling reaction and a diastereoselective Sm(II)-mediated reductive cascade cyclization reaction. These transformations enabled the preparation of (-)-maoecrystal Z in only 12 steps from (-)-γ-cyclogeraniol.
The enzyme-based system performing a biocatalytic cascade reaction was realized in a flow device and was used to mimic Boolean logic operations. Chemical inputs applied to the system resulted in the activation of additional reaction steps, allowing the reversible switch of the logic operations between OR, NXOR and NAND gates for processing of two other biomolecular inputs.
