A 3D microvascularized gelatin hydrogel is produced using thermoresponsive sacrificial poly(N-isopropylacrylamide) microfibers. The capillary-like microvascular network allows constant perfusion of media throughout the thick hydrogel, and significantly improves the viability of human neonatal dermal fibroblasts encapsulated within the gel at a high density.

Current cell sheet-based blood vessels lack biomimetic structure and require excessively long culture times that may compromise smooth muscle cell phenotype. We modified a commercially available product for uniaxial cell sheet conditioning with thermoresponsive copolymers. Thus, culture of detachable conditioned cell sheets is shortened while retaining structural integrity and contractility.

A synthetic pathway is described to construct thermoresponsive freestanding nanomembranes at the aqueous-air interface of a pendant drop. Dynamic control of the reaction kinetics allows formation of viscoelastic interfaces supporting anisotropic stresses and mechanical stability, which can be tuned by external stimuli.
