originated (data not shown), one would perhaps expect to see mutations being lost or gained within the genes potentially affecting quinolone susceptibility. The stability of mutations in genes known to be associated with fluoroquinolone resistance, and the stable MIC values in clonally related MRSA isolates over time, have important implications for quinolone resistance in S. aureus. From this ...

Bacteria develop resistance to antimicrobial agents by a number of different mechanisms. The resistance to (fluoro)quinolones in Salmonella is of particular importance especially if therapy in humans is required. For decades there has been a significant interest in studying the biology of Salmonella because these bacteria are among the leading causes of foodborne illnesses around the globe. To ...

BACKGROUND The use of quinolone prophylaxis in high-risk neutropenic patients is considered standard of care but the development of resistance is a concern. Previous studies have focused mainly on quinolone resistance among patients receiving prophylaxis, with very few data reporting its impact on the hospital microbial epidemiology. METHODS We analyzed a cohort of 329 episodes of chemotherap...

background: emergence klebsiella pneumoniae resistant to quinolone antibiotics due to mutations in gyra and parc genes created problem for treatment of patients in different hospitals in iran. the objective of this study was to determine the amino acid substitutions of gyra and parc proteins in certain clonal lineages of the k. pneumoniae conferring high level quinolone resistance. methods: one...

AIMS The aims of this study were to investigate the epidemiology of quinolone-resistant and -susceptible porcine isolates of Campylobacter coli and to characterize the genetic basis of quinolone resistance. METHODS AND RESULTS Penner serotyping and flagellin gene sequence polymorphisms were used to investigate the epidemiology of the C. coli isolates. A total of 55 isolates were included, of ...

BMS-284756, a novel des-fluoro(6)-quinolone, was used to select for in vitro mutants of Staphylococcus aureus ISP794. Step mutants were obtained, and the quinolone resistance-determining regions of four target genes, gyrA, gyrB, grlA, and grlB, were sequenced. The data suggest that DNA gyrase is the primary target for BMS-284756 in S. aureus.

We developed a novel rapid assay to detect the gyrA mutations that cause quinolone resistance in typhoid and paratyphoid fever Salmonella spp. using high-resolution melting (Idaho Technology, Salt Lake City, UT) analysis of polymerase chain reaction amplicons. The presence of gyrA mutations led to small but consistent changes in amplicon melting temperatures that allowed quinolone-resistant iso...

The uptakes of norfloxacin by quinolone-resistant and -susceptible strains of Serratia marcescens were almost the same and 50% inhibitory concentrations for DNA gyrase and the MICs of quinolones were correlated, suggesting that DNA gyrase alterations are the basis of quinolone resistance.

Binding of the quinolone drug norfloxacin to gyrase and DNA has been investigated. We have detected binding to gyrase-DNA complex but find no significant binding to either gyrase or DNA alone. Enzyme containing gyrase A protein with the mutation Ser-83 to Trp (conferring quinolone resistance) showed greatly reduced drug binding.

To evaluate the molecular mechanism of fluoroquinolones resistance in Mycoplasma hominis (MH) clinical strains isolated from urogenital specimens. 15 MH clinical isolates with different phenotypes of resistance to fluoroquinolones antibiotics were screened for mutations in the quinolone resistance-determining regions (QRDRs) of DNA gyrase (gyrA and gyrB) and topoisomerase IV (parC and parE) in ...
