Background: The B-cell antigen CD20 is expressed on normal B-cells and almost all lymphomas. This non-modulating agent provides an excellent target for antibody-directed therapeutic regimens. IDECC2B8 (rituximab) a monoclonal antibody (mAb) directed against the B-cell-specific CD20. mediates complement antibody-dependent cell-mediated cytotoxicity. As its usage has surged, there have been growi...

Background: The antibody response to hepatitis B surface antigen (HBsAg) controls hepatitis B virus infection. The "a" determinant of HBsAg is the most important target for protective antibody response, diagnosis and immunoprophylaxis. Mutations in this area may induce immune escape mutants and affect the performance of HBsAg assays. Objectives: To construct clinically relevant recombinant muta...

Nine hybridomas secreting monoclonal antibody to proteins of bovine milk-fat-globule membrane were isolated. All nine cell lines continued to secrete monoclonal antibody after serial transfer in culture and after passage as solid tumours in Balb/cJ mice. Four of the cell lines secreted monoclonal antibody specific for xanthine oxidase, one of the major proteins of milk-fat-globule membrane.

Analysis of human fibrosarcoma cells exposed to radiolabeled monoclonal antibody 19-24, which recognizes sarcoma-associated antigen p102, revealed that over 54% of the cell surface-bound radioactivity was internalized. No modulation of cell surface p102 antigen by monoclonal antibody 19-24 was observed in human fibrosarcoma cells. Monoclonal antibody 19-24 coupled to daunomycin via a dextran br...

Background: Sjögren’s syndrome is the second most common systemic autoimmune disease after rheumatoid arthritis, RA, with a prevalence of about 0.5% in the general population. It occurs primarily in perimenopausal women (at a ratio of women to men of 9: 1) Sjögren’s syndrome is characterized by lymphocytic infiltration to exocrine glands. The specific autoantibodies of this syndrome are against...

Background: It is believed that the loading value of anticancer drug conjugated to the monoclonal antibody, called drug-to-antibody ratio (DAR), is the main quality feature of antibody-drug conjugates. Methods: In this study, matrix assisted laser desorption/ionization mass spectrometry was used to determine the average molecular weight of trastuzumab and its three conjugated forms. The differe...

Evolution of therapeutic monoclonal antibodies significantly benefited from the recombinant DNA technologies that are used to generate chimeric, humanized, and human versions of monoclonal antibody to reduce the problem of immunogenicity and neutralization, as well as from understanding mechanisms of action mediated by monoclonal antibodies (Nelson et al., 2010; Reichert, 2009; Ranson & Sliwkow...

Plasma exchange was used to treat 10 patients with polyneuropathy and a monoclonal antibody (plasma cell dyscrasia). Six patients had improvement of the neuropathy, while three patients had stabilisation of the neuropathy during plasma exchange. The patients who improved maintained a 64% or greater decrease in the monoclonal antibody between exchanges. Patients with axonal polyneuropathy as wel...
