APOBEC3G catalyzes deamination of cytosines in HIV-1 genome, and restricts the HIV-1 infection. Here, we propose a picomole-scale assay for the detection of DNA deamination catalyzed by APOBEC3G. Our results show the suitability of the developed method for a time course analysis of enzyme-catalyzed DNA modifications.

The deamination of adenosine by various tissue preparations was studied by Schmidt in 1928 and later by Klein (1941), and Conway and Cooke (1939). McElroy and Mitchell (1946) demonstrated the presence of the enzyme in Neurospora and in Aspergilli. The enzyme was partially purified by Kalckar (1947) in a series of studies on purine-related enzymes in calf mucosa homogenate. Recent miscellaneous ...

Histidase catalyzes the irreversible nonoxidative deamination of histidine to urocanic acid. This reaction (Equation 1) has been studied in mammalian and bacterial preparations (1). Although enzymes which carry out similar deaminations of other amino acids (notably aspartic acid) have been described, to date no detailed studies of the mechanism of the process of nonoxidative deamination have be...

Stopped flow studies of D2O kinetic solvent isotope effects on the reaction catalyzed by L-glutamate dehydrogenase reveal, in addition to several effects apparently attributable simply to pKa shifts, a 2-fold pH-independent effect on the velocity of the steady state oxidative deamination of L-glutamate by enzyme and NADP. Comparable pH-independent D2O kinetic solvent isotope effects are seen bo...

PSYCHIATRIC ANNALS • Vol. 44, No. 11, 2014 495 ABSTRACT Monoamine oxidase (MAO) isoenzymes A and B are mitochondrial-bound proteins that catalyze the oxidative deamination of dietary amines and monoamine transmitters. Others include: serotonin, epinephrine, 2-phenylethylamine, and dopamine. MAOs can potentially modulate all the processes involving bioactive amines, including regulation of mood,...

Three hundred bacterial isolates from soil were tested for resistance against phosphinothricin [PPT; dl-homoalanin-4-yl(methyl)phosphinic acid], the active ingredient of the herbicide BASTA. Eight resistant bacterial strains and Escherichia coli were analyzed for PPT-transforming activities. At least three different enzymatic reactions could be detected in cell extracts. In six strains an acety...

Pyridoxal 5'-phosphate (PLP)-dependent enzymes have wide catalytic versatility but are rarely known for their ability to react with oxygen to catalyze challenging reactions. Here, using in vitro reconstitution and kinetic analysis, we report that the indolmycin biosynthetic enzyme Ind4, from Streptomyces griseus ATCC 12648, is an unprecedented O2- and PLP-dependent enzyme that carries out a fou...

Stopped flow studies of the oxidative deamination of L-glutamate by TPN and beef liver glutamate dehydrogenase were performed at pH 6.5 and 7.6. At each pH value, the initial burst slopes obey an equation of the form: e/v = 40 + dd(TPN) + h/bglutamate) + &/(TPN)(L-glutamate). The agreement of dissociation constants for enzyme-TPN and enzyme-L-glutamate binary complexes obtained from the r#~ val...
