T h e processes of DNA replication, DNA ligation, and RNA transcription have provided the basis of distinct and separate in vitro nucleic acid amplification strategies. The first described and the most widely utilized amplification protocol is the polymerase chain reaction (PCR).O,2) The PCR method consists of multiple cycles of oligonucleotide primer-directed DNA replication that are punctuate...

background: baculoviral inhibitor of apoptosis repeat-containing 5 (birc5) gene is an inhibitor of apoptosis that expresses in human embryonic tissues but it is absent in most healthy adult tissues. the copy number of birc5 has been indicated to be highly increased in tumor tissues; however, its association with the age of onset in breast cancer is not well understood. methods: forty tumor tiss...

A novel DNA assay is demonstrated which relies on the production of a surface enhanced resonance Raman scattering (SERS) signal following sequence specific hybridisation and enzyme hydrolysis. This method achieves high sensitivity and specificity that can be applied to the detection of unlabelled duplex DNA and is the first step towards a SERS based signal amplification approach.

With global efforts to scale up the prevention of mother-to-child transmission services and pediatric antiretroviral therapy, there is an urgent need to introduce a simple, low-cost infant human immunodeficiency virus test in the field. We postulated that the p24 antigen capture enzyme-linked immunosorbent assay could be simplified by eliminating signal amplification without compromising diagno...

Enzymatic recognition of unlocked nucleic acid (UNA) nucleotides was successfully accomplished. Therminator DNA polymerase was found to be an efficient enzyme in primer extension reactions. Polymerase chain reaction (PCR) amplification of a 81 mer UNA-modified DNA library was efficiently achieved by KOD DNA polymerase.

A novel fluorescent nanosensor has been developed by combining super fluorescence quenching ability of graphene oxide and hybridization chain reaction amplification, which enables highly sensitive detection of base excision repair enzyme activity with a wide dynamic range from 0.0001 to 100 U mL(-1) and a detection limit of 0.00006 U mL(-1).

A comparative study of herpes simplex virus diagnosis by standard cell culture and a new hybrid test (enzyme-linked immunosorbent assay spin amplification technique) was done on 300 specimens. The new test was found to be equally sensitive and specific, much less expensive to perform, and to report all results in 48 h.

Recent studies have revealed that multiple intracellular signaling proteins may assemble into structured, yet sometimes infinite, higher-order signaling machines for transmission of receptor activation information to cellular responses. These studies advance our understanding of cell signaling and implicate new molecular mechanisms in proximity-driven enzyme activation, threshold behavior, sign...
