This application note describes how the Agilent Bio-Monolith Protein A column was applied to the optimization of Chinese hamster ovary cell-culture conditions to produce a recombinant monoclonal antibody with desired structural characteristics, focusing on glycosylation. The workflow demonstrates the versatility and wide applicability of the column in biopharmaceutical and biosimilar development.

Mammalian cells dominate the biopharmaceutical industry and maintaining cell viability is a limiting factor in most mammalian cell cultures. This project is focused on developing an online (in-situ) dielectric spectroscopy sensor that is capable of measuring cell viability in real-time to enable optimization of culture conditions for Chinese Hamster Ovary (CHO) cell lines in micro-bioreactors.

Three transfection reagents, Lipofectamine 2000, TransIT-PRO and linear 25 kDa polyethylenimine were evaluated for transient expression of enhanced green fluorescent protein in Chinese hamster ovary cells. TransIT-PRO was found to be more efficient under the examined conditions, but comes at an increased cost compared to the widely used PEI.

Chinese hamster ovary (CHO) cells are the primary host used for biopharmaceutical protein production. The engineering of CHO cells to produce higher amounts of biopharmaceuticals has been highly dependent on empirical approaches, but recent high-throughput "omics" methods are changing the situation in a rational manner. Omics data analyses using gene expression or metabolite profiling make it p...

BACKGROUND The optimization of bioprocess conditions towards improved growth profile and productivity yield is considered of great importance in biopharmaceutical manufacturing. Peptones as efficient sources of nutrients have been studied for their effect on media development; however, their role on metabolic pathway is not well understood. METHODS In the present study, the effect of differen...

MicroRNAs (miRNAs) are small non-coding RNAs (∼22 nucleotides) which regulate gene expression by silencing mRNA translation. MiRNAs are transcribed as long primary transcripts, which are enzymatically processed by Drosha/Dgcr8, in the nucleus, and by Dicer in the cytoplasm, into mature miRNAs. The importance of miRNAs for coordinated gene expression is commonly accepted. Consequentially, there ...

High throughput (HT) platforms serve as a cost-efficient and rapid screening method for evaluating the effect of cell-culture conditions and screening of chemicals. We report the development of a HT cell-based microarray platform to assess the effect of culture conditions on Chinese hamster ovary (CHO) cells. Specifically, growth, transgene expression and metabolism of a GS/methionine sulphoxim...

We report the synthesis and radioligand binding analysis of a series of naphthalenic melatonin receptor ligands, N-[2-(7-alkoxy-2-methoxy-1-naphthyl)ethyl]propionamide. This series of ligands exhibits subpicomolar binding affinity to both MT1 and MT2 melatonin receptors expressed in chinese hamster ovary (CHO) cells.

The findin«that 7r,8f-dihydroxy-9,10f-oxy-7,8,9,10-tetrahydrobenzo(a)pyrene (BPDE-I) is stabilized against hydrolysis by binding to cellular membranes suggested that nucleophilic compounds which would colocalize with BPDE-I in membranes might inhibit the deleterious biological effects of BPDE-I. We have explored the possibility that hydrophobic, sulfhydryl-containing compounds might provide su...
