The variability and adaptability of the amoebae from the class Dictyosteliomycetes greatly complicate their systematics. The nucleotide sequences of the ribosomal internal transcribed spacers and the 5.8S ribosomal DNA gene have been determined for 28 isolates, and their utility to discriminate between different species and genera has been shown.

The aim of the present study was to analyse sequences of 16S-23S internal transcribed spacer regions of different Geobacillus species, identify structural elements of these regions and evaluate the possibility to use these structural elements in the taxonomy of the genus Geobacillus. GEOBAC-PCR was used in order to obtain 16S-23S rRNA internal transcribed spacer sequences. Certain structural el...

Sequences of the internal transcribed spacers (ITS) of the nuclear ribosomal DNA are important molecular markers in phylogenetic analyses. To obtain sequences from herbarium material in which DNA often is severely degraded, the ITS region has to be amplified in two steps. Two methods that reduce bench time and reagents used are described. (i) Separately amplified preparations of subunits ITS-1 ...

Symptomatic samples of anthracnose-twister disease cultivated onion (Allium cepa) were collected from heavily infected fields at Matale district, Sri Lanka during a severe outbreak that occurred through late July to August in 2020. The pathogen was identified as Colletotrichum siamense based on morphology and combined gene sequence analysis nuclear ribosomal internal transcribed spacers 1 2 wit...

Nuclear ribosomal sequence data from the internal transcribed spacers (ITS-1 and ITS-2), 5.8S subunit, and regions of the 18S and 28S genes were used to investigate sequence diversity among geographic samples of Nematodirus battus, and to infer phylogenetic relationships among Nematodirus species. Phylogenetic analysis of these data yielded strong support for relationships among species, depict...

Sampling of agricultural soils from the Mexican northeastern region was performed to detect Trichoderma spp., genetically characterize it, and assess its potential use as a biologic control agent against Macrophomina phaseolina. M. phaseolina is a phytopathogen that attacks over 500 species of cultivated plants and causes heavy losses in the regional sorghum crop. Sampling was performed immedia...

The Puccinia monoica complex is an enigmatic group of rust fungi. They are flower mimics, and they greatly reduce host reproduction and survival. These fungi are relatively common, attacking approximately 960 species in 11 genera of crucifers as well as at least five genera of grasses. In modern taxonomic treatments the Puccinia monoica complex is treated as four species that are differentiated...

Phylogenetic relationships among 28 of the 68 species of the paleotropical genus Gaertnera (Rubiaceae) and two related genera were inferred from nucleotide sequence variation in four nuclear DNA (nDNA) markers: the internal transcribed spacers of nuclear rDNA (ITS), the large and small copies of phosphoenolpyruvate carboxylase (PepC-large and PepC-small), and triose phosphate isomerase (Tpi). P...

Sooty blotch and flyspeck occur on fruit surfaces and result in economic losses due to less attractive appearance. Sooty blotch fungi form dark mycelial mats whereas flyspeck fungal agents are well characterized with black, sclerotium-like bodies on fruit surface. Whilst more than 60 species have been reported in association with these two fungal diseases, thus far they have not been studied in...

Restriction fragment length polymorphism (RFLP) analysis was performed on the internal transcribed spacer regions of 204 Sporothrix schenckii isolates and on one strain each of the related fungi, S. schenckii var. luriei, S. curviconia, S. inflata and Ceratocystis stenoceras. S. schenckii isolates, which have been collected from around the world, have already been typed according to their mitoc...
