BACKGROUND/AIMS Insulin-like growth factor-1 receptor (IGF-1R) plays critical roles in cell proliferation, differentiation, apoptosis, and transformation. Suppression of IGF-1R by means of antisense methods and specific antibodies causes cell apoptosis and growth inhibition of cancer cells. The present study aims to investigate whether there is a difference between normal and cancerous tissue w...

This report reviews the current knowledge on the biosynthesis, regulation and secretion of Growth Hormone (GH) and Insulin-like Growth factor-I (IGF-I). Variants of human GH, molecular size variants as well as charge variants, and the GH binding protein will be discussed, and the regulation of expression of the hGH gene is reviewed. IGF-I expression, regulation of the igf-I gene and secretion i...

Melanocytes, highly differentiated neural crest-derived cells, are located in the basal layer of the epidermis, where they play a role in protecting against UV damage in the skin. Previous studies suggest that both growth hormone (GH) and the insulin-like growth factor I (GH/IGF-I) system may be important for melanocyte growth and function. We have therefore characterized the role of the GH/IGF...

UNLABELLED Poor postnatal growth after preterm birth does not match the normal rapid growth in utero and is associated with preterm morbidities. Insulin-like growth factor 1 (IGF-1) axis is the major hormonal mediator of growth in utero, and levels of IGF-1 are often very low after preterm birth. We reviewed the role of IGF-1 in foetal development and the corresponding preterm perinatal period ...

Insulin receptor substrate-1 (Irs1) is one of the major substrates for insulin and insulin-like growth factor-1 (IGF-1) tyrosine kinases. Systemic Irs1-deficient mice show retardation, with resistance to IGF-1, although underlying mechanisms remain poorly understood. For this study, we generated brain-specific deletion Irs1 (NIrs1KO mice). The NIrs1KO exhibited lower body weights, shorter bodie...

Insulin and insulin-like growth factors I and II are closely related protein hormones. Their distinct evolution has resulted in different yet overlapping biological functions with insulin becoming a key regulator of metabolism, whereas insulin-like growth factors (IGF)-I/II are major growth factors. Insulin and IGFs cross-bind with different affinities to closely related insulin receptor isofor...

analysis showed that this was a splice variant of the insulin-like growth factor-I (IGF-I) gene, although its 3 exons were different from the liver or systemic type of IGF-I. However, it was noted that muscle also expresses the systemic type of IGF-I (IGF-IEa). The terminology for the IGF-I variants is a problem when attempting to apply it to nonhepatic tissues and to different species. Also, w...

Specific, high affinity insulin-like growth factor (IGF) binding proteins are secreted by human fibroblasts in culture. By multiple criteria, the species of IGF binding proteins produced by human fibroblasts are distinct from the HepG2/amniotic fluid IGF binding protein, but share many characteristics with the growth hormone-dependent IGF binding protein forms predominant in normal adult human ...

Short stature is a characteristic feature of pycnodysostosis. We report defective growth hormone secretion in response to provocation and low insulin-like growth factor-I (IGF-I) concentration in five out of six patients with pycnodysostosis. Physiological replacement with growth hormone increased IGF-I concentration and improved linear growth in these children.
