Hybrid micro-/nano-silica architectures made of bacterial-body-templated silica shell attached with fimbriae-templated silica nanotubes were prepared by using living fimbriaed bacteria E. Coli TG1 as templates, which can function as individual bioreactors to catalyze the hydrolysis of 4-nitrophenyl-beta-d-galactopyranoside to form p-nitrophenol due to the presence of an intracellular enzyme.

Herein, we report the synthesis of 2nd generation PPDs functionalized with free thiol moieties within the scaffold, which were used as anchor points for the covalent attachment of guest species (p-nitrophenol derivatives) through the oxidative formation of disulfide linkages. The disulfide bonds were then cleaved under reductive conditions using dithiothreitol to discharge the molecules.

A degradative bacterium, M6, was isolated and presumptively identified as Plesiomonas sp. strain M6 was able to hydrolyze methyl parathion to p-nitrophenol. A novel organophosphate hydrolase gene designated mpd was selected from its genomic library prepared by shotgun cloning. The nucleotide sequence of the mpd gene was determined. The gene could be effectively expressed in Escherichia coli.
