In order to investigate the relationship between depth within membranes of singlet oxygen generation and effectiveness of photodynamic therapy of tumors, analogs of protoporphyrin-IX 1 bearing five 4 and seven 5 carbon atoms (in place of the 3-carbon atom chain in 1) were synthesized from monopyrrole precursors.

Photosensitizers, Zn protoporphyrin IX and Zn chlorin e6, are completely inserted into each heme pocket of a hexameric apohemoprotein. The fluorescence quenching efficiencies upon addition of methyl viologen are 2.3 and 2.6 fold-higher than those of the partially photosensitizer-inserted proteins, respectively, indicating that the energy migration occurs within the proteins.
Comparison of the collision cross-section of the [Fe(III)-protoporphyrin IX]+ ion, hemin+, measured by means of ion-mobility experiments and the cross-sections calculated from theoretical structures based on density functional theory reveals that hemin+, in the gas phase, contains intramolecular hydrogen bonding between its two propionic acid side-chains.
