In 1902, Charles Richet coined the term “anaphylaxis” to describe a “state of heightened sensitivity subject substance induced by first injection, that instead protecting organism, renders it more fragile and susceptible.” Since this description, experimental work led identification antibodies, receptors, cells, mediators in severe allergic reaction, leading paradigm anaphylaxis is an IgE-depen...

Distinct genes encode 6 human receptors for IgG (hFcgammaRs), 3 of which have 2 or 3 polymorphic variants. The specificity and affinity of individual hFcgammaRs for the 4 human IgG subclasses is unknown. This information is critical for antibody-based immunotherapy which has been increasingly used in the clinics. We investigated the binding of polyclonal and monoclonal IgG1, IgG2, IgG3, and IgG...

The mononuclear and polymorphonuclear cells of the body have the capacity to bind, phagocytose, and ultimately destroy antibody-coated particles such as bacteria by a series oflinked molecular processes that initially involve the plasma membrane Fc receptors for IgG (FcyR) . Our understanding of these FcyR-mediated processes is rudimentary. We know that first the Ig-coated particles bind to the...

The middle part of the heavy chain of IgG3 (hinge region) which covalently links the two y3 chains to each other, is about 4 times larger than the same region in the three other human IgG subclasses. This is probably due to a quadruplication of a 45-nucleotide DNA segment resulting in a y3 hinge region which is 62 amino acid residues long and consists of an NH,-terminal 17-residue segment follo...

BACKGROUND Antigen specificity and IgG subclass could be significant in the natural history of Chagas' disease. The relationship between the different stages of human Chagas' disease and the profiles of total IgG and its subclasses were thus analysed here; they were directed against a crude T. cruzi extract and three recombinant antigens: the T. cruzi kinetoplastid membrane protein-11 (rKMP-11)...

Analysis of mouse IgG binding to Fc receptors on mouse B cells indicates that the IgG1, IgG2a, and IgGb subclasses bind to the same receptor. No differences in affinity were detected among subclass or between mouse strains. This same receptor bound rat IgG with an affinity that differed between mouse strains. This polymorphism in affinity for rat IgG maps to chromosome 12 distal to the Igh locus.

Cerebral toxoplasmosis can be highly debilitating and occasionally fatal in persons with immune system deficiencies. In this study, we evaluated the Toxoplasma gondii-specific IgG subclass antibody response in 19 cerebrospinal fluid (CSF) samples from patients with cerebral toxoplasmosis who had a positive IgG anti-T. gondii ELISA standardized with a cyst antigen preparation. There were no sign...

Polyclonal anti-immunoglobulin G (anti-IgG) secondary antibodies are essential tools for many molecular biology techniques and diagnostic tests. Their animal-based production is, however, a major ethical problem. Here, we introduce a sustainable alternative, namely nanobodies against all mouse IgG subclasses and rabbit IgG. They can be produced at large scale in Escherichia coli and could thus ...
