Cytochalasin-induced actin disruption has often been associated with decreased bacterial internalization by cultured epithelial cells, although polarized enterocytes have not been systematically studied. In assays using confluent polarized HT-29 enterocytes, cytochalasin D appeared to increase internalization of wild-type Salmonella typhimurium, Proteus mirabilis, and Escherichia coli. HeLa and...

Intestinal transporter proteins and metabolizing enzymes play a crucial role in the oral absorption of a wide variety of drugs. The aim of the current study was to characterize better available intestinal in vitro models by comparing expression levels of these proteins and enzymes between porcine intestine, human intestine, and Caco-2 cells. We therefore determined the absolute protein expressi...

Human colon carcinoma Caco-2 cell monolayers undergo conversion into cells that share morphological and functional features of M cells when allowed to interact with B lymphocytes. A lymphotropic (X4) HIV-1 strain crosses M cell monolayers and infects underlying CD4(+) target cells. Transport requires both lactosyl cerebroside and CXCR4 receptors, which are expressed on the apical surface of Cac...

Studies with human neonatal rotaviruses RV-3 and S12/85 and their reassortants showed that VP4 is a determinant of rotavirus attachment to and growth in Caco-2 cells. The binding of these viruses to MA104 and Caco-2 cells correlated with their growth ability. Virus sensitivity to trypsin and the VP4 fusion region may be implicated in these processes.

The Caco-2 cell line and its sub-clone TC7 are frequently used for studying human intestinal transport and metabolism of xenobiotics. We have investigated the expression of soluble sulfotransferases (SULT) in parental Caco-2 and TC7 cells by immunoblotting. SULT1A1, 1A2, 1A3, 1B1, 1C1, 1C2 and 2A1 were expressed in both cell lines. SULT2B1a, 2B1b and 4A1 were absent. SULT1E1 protein was found i...

The present study used various amounts of P-arsanilic acid (AsA) in pot experiments to evaluate the effects AsA on arsenic (As) accumulation, speciation and meanwhile using vitro digestion/Caco-2 cell model bioavailability As rice. results indicated a linear relationship between rice soil, at 75 mg AsA/kg content exceeded statutory permissible limit 0.2 As/kg dry weight China. Speciation studie...

Lactoferrin (LF) is an iron-binding glycoprotein contained in milk and other exocrine fluids, and is believed to have multiple biological functions. We investigated the intracellular dynamics of LF taken up by three lines of human enterocytes and the subsequent release of internalized LF by using two-site ELISA and confocal microscopy. LF taken up by Caco-2 cells was kept partially intact withi...

A method for the detection of infectious human rotaviruses based on infection of CaCo-2 cells and detection of infected cells by indirect immunofluorescence and flow cytometry (IIF-FC) has been developed. The technique was validated by performing a seminested reverse transcription-PCR assay with sorted cell populations. The efficiency of the procedure has been compared with that of the standard...

Drug delivery by folate conjugated amphiphiles Abstract A series of amphiphilic conjugates of folic acid and aliphatic fatty acids was designed to serve as targeted drug delivery carriers for anticancer agents. The effectiveness of targeted delivery of amphiphile was evaluated using folate receptor positive HeLa cells and Caco-2 cells that do not express folate receptor. Wild type HeLa cells we...

Background: Tight junctions are found in epithelial cells and function as selective gatekeepers to regulate absorption. PT-gliadin is the gluten protein segment that is known to impair the functioning of tight junctions. This study aimed to examine the effects of a lignite extract dietary supplement (RESTORE) on tight junction function in small intestine (IEC-6) and colon (Caco-2) epithelial ce...
