background and objective: acinetobacter baumannii is an aerobic non-motile gram-negative bacterial pathogen that is resistant to most antibiotics. carbapenems are the most common antibiotics for the treatment of infections caused by this pathogen. mechanisms of antibiotic-resistance in a. baumannii are mainly mediated by efflux pumps-lactamases. the aim of this study was to determine antibiotic...

a modified pulsed-field gel electrophoresis (pfge) protocol was developed and applied to clinical isolates of mycobacterium tuberculosis complex to reduce the cost of using lyticase. this protocol reduces the expense of pfge typing of mycobacterium tuberculosis complex as it removes the use of lyticase during the spheroplast formation from these bacteria.

conclusions molecular epidemiology studies of infectious diseases have been useful when bacterial isolates have been clustered in a period of time and in different geographical regions with variable antibiotic resistance patterns. in spite of high geographical differences and different antibiotic resistant patterns, low genetic diversity among the studied tb isolates may refer to the low rate o...

PURPOSE The purpose of this work was to examine the tear film proteome using a combination of one-dimensional (1D) and two dimensional (2D) gel electrophoresis and mass spectrometry-based techniques and to explore the effect of the tear collection methods on the tear proteome. METHODS Tear samples from eight normal non-contact lens wearing human subjects collected by Drummond glass microcapil...

Post-translational modification expands the functionality of the proteome beyond genetic encoding, impacting many cellular processes. Cleavage of the carboxyl terminus is one of the many different ways proteins can be modified for functionality. Gel-electrophoresis and mass spectrometric-based techniques were used to identify proteins impacted by deficiency of a C-terminal protease, CtpA, in Rh...

Standardized, comprehensive platforms for the discovery of protease substrates have been extremely difficult to create. Screens for protease specificity are now frequently based on the cleavage patterns of peptide substrates, which contain small recognition motifs that are required for the cleavage of the scissile bond within an active site. However, these studies do not identify in vivo substr...

Despite extensive study, progress in elucidation of biological functions of amyloids and their role in pathology is largely restrained due to the lack of universal and reliable biochemical methods for their discovery. All biochemical methods developed so far allowed only identification of glutamine/asparagine-rich amyloid-forming proteins or proteins comprising amyloids that form large deposits...

We propose a statistical model-based approach for the segmentation of fragments of DNA as a first step in the automation of the primarily manual process of comparing two or more images resulting from the Restriction Landmark Genomic Scanning (RLGS) method. These 2D gel electrophoresis images are the product of the separation of DNA into fragments that appear as spots on X-ray films. The goal is...

We used 2D protein gel electrophoresis and DNA microarray technologies to systematically analyze genes under glucose repression in B:acillus subtilis. In particular, we focused on genes expressed after the shift from glycolytic to gluconeogenic at the middle logarithmic phase of growth in a nutrient sporulation medium, which remained repressed by the addition of glucose. We also examined whethe...

We have investigated the plasma proteome by using 2D gel electrophoresis and MS from patients with severe acute respiratory syndrome (SARS). A complete proteomic analysis was performed on four patients with SARS in different time courses, and a total of 38 differential spots were selected for protein identification. Most of the proteins identified are acute phase proteins, and their presence re...
