A general procedure, involving 4-dimethylaminopyridine-catalysed acylation of the amino groups in the appropriate S-blocked glutathione, is reported for the preparation of N-blocked derivatives of glutathiones.

Additions of lithium phenylacetylide to a Weinreb amide are described. Dimeric lithium acetylide reacts via a monosolvated monomer-based transition structure. The first total synthesis of monosporascone is presented. The five-step synthesis developed includes a silver acetylide-acid chloride coupling, domino Diels-Alder-retro-Diels-Alder reaction, and an intramolecular Friedel-Crafts acylation, and provides the natural product in 57% yield overall. Selective reduction of monosporascone also afforded the related metabolite dihydromonosporascone.

Vinyl and p-nitrophenyl esters have emerged as alternative substrates for the LovD9 acylation mechanism, showing product yields high those obtained by α-dimethylbutyryl-S-methyl-3-mercaptopropionate (DMB-SMMP), thioester which was evolved.
