Mass spectrometry is a selective and powerful technique to obtain identification and structural information on compounds present in complex mixtures. Since it requires only small sample amount it is an excellent tool for researchers interested in detecting changes in composition of complex carbohydrates of plants. This mini-review gives an overview of common mass spectrometry techniques applied...

Cell walls are crucial for development, signal transduction, and disease resistance in plants. Cell walls are made of cellulose, hemicelluloses, and pectins. Xyloglucan (XG), the principal load-bearing hemicellulose of dicotyledonous plants, has a terminal fucosyl residue. A 60-kilodalton fucosyltransferase (FTase) that adds this residue was purified from pea epicotyls. Peptide sequence informa...

BACKGROUND Understanding the biological mechanisms used by microorganisms for plant biomass degradation is of considerable biotechnological interest. Despite of the growing number of sequenced (meta)genomes of plant biomass-degrading microbes, there is currently no technique for the systematic determination of the genomic components of this process from these data. RESULTS We describe a compu...

How is the extensibility of growing plant cell walls regulated? In the past, most studies have focused on the role of the cellulose/xyloglucan network and the enigmatic wall-loosening agents expansins. Here we review first how in the closest relatives of the land plants, the Charophycean algae, cell wall synthesis is coupled to cell wall extensibility by a chemical Ca(2+)-exchange mechanism bet...

Pectins isolated from sugar beet pulp by autoclaving contained significant amounts of ferulates, 8.8% of which were ferulate dehydrodimers. The 8-8 and 8-0-4 dehydrodimers were predominant. Oxidative cross-linking with hydrogen peroxide/peroxidase lowered the amount of ferulic acid by 78%, while an increase in ferulate dehydrodimers by a factor of 4.9 was observed. The highest increase was seen...

Peaches (Prunus persica cv. Hermoza) were stored at 0C in regular air (RA) or in controlled atmosphere (CA 10% CO2, 3% O2) for 4 weeks and then ripened for 4 days at 20 degrees C. Woolliness developed in the regular air stored fruit while the controlled atmosphere stored fruit ripened normally. In the woolly fruit symptoms of the disorder were greater in the inner mesocarp than in the outer. Po...
