A fluorimetric assay for monitoring depurination of the N7-alkylguanine adduct derived from the anticancer natural product leinamycin is described. This general approach could potentially provide the foundation for a high throughput assay that detects DNA-alkylating agents or a convenient continuous fluorimetric assay for base excision repair enzymes.

A sensitive fluorimetric method was developed for the quantitative determination of DNA in plant (Zea mays L. and Medicago sativa L.) extracts. This method takes advantage of the specific increase in fluorescence intensity of the complex of DNA and the dye 4',6'-diamidino-2-phenylindole (DAPI). Recovery of DNA and dissociation of histones from DNA were maximized by the addition of 2.0 molar NaC...

A sensitive fluorimetric assay based on DNA binding of the fluorescent dye Hoechst 33258 was developed to quantitate cell numbers in microtiter plates. The cells were fixed and incubated with the dye solution. The bound dye was extracted with ethanol and fluorescence was measured in a spectrofluorimeter. The method is sensitive for as little as a thousand cells and is particularly useful for th...

Since the observation of Desha (1) that the fluorescence exhibited by solutions of quinine and aniline erythrosine when irradiated by ultraviolet light could be utilized for their quantitative determination, useful fluorimetric methods have been devised for a number of biological constituents (2). The extreme sensitivity of fluorimetric assay offers a distinct advantage over other chemical and ...

BACKGROUND Biotin, a water-soluble vitamin, is used as a co-factor by enzymes involved in carboxylation reactions. Biotinidase (BTD) catalyzes the recycling of biotin from endogenous and dietary sources. Biotinidase deficiency (BD) is an autosomal recessively inherited disorder of biotin recycling that is associated with neurologic and cutaneous consequences when untreated. The aim of the study...

A fluorimetric microassay that uses a redox dye to determine the viability of the flagellate Trichomonas vaginalis has been optimised to provide a more sensitive method to evaluate potential trichomonacidal compounds. Resazurin has been used in recent years to test drugs against different parasites, including trichomonadid protozoa; however, the reproducibility of these resazurin-based methods ...

cells, proteins, and small molecules from the alveolar space of the lower respiratory tract since the first report of Reynolds and Newball over three decades ago. The analysis of these components is of wide diagnostic and investigative value. In bronchoalveolar lavage, sterile saline is administered into the bronchoalveolar space and aspirated by syringe. Because recovery of the components in b...
