We present a sensitive colorimetric determination of 5'-nucleotidase based on the measurement of liberated phosphate by reaction with stannous chloride/molybdenum blue. No protein precipitation is required. The method is simple, sensitive, economical, and easy to use, and involves relatively stable reagents.

The first purification of the Mo-reducing enzyme from Serratia sp. strain DRY5 that is responsible for molybdenum reduction to molybdenum blue in the bacterium is reported. The monomeric enzyme has an apparent molecular weight of 105 kDalton. The isoelectric point of this enzyme was 7.55. The enzyme has an optimum pH of 6.0 and maximum activity between 25 and 35°C. The Mo-reducing enzyme was ex...

Bacteria with the ability to tolerate, remove, and/or degrade several xenobiotics simultaneously are urgently needed for remediation of polluted sites. A previously isolated bacterium with sodium dodecyl sulfate- (SDS-) degrading capacity was found to be able to reduce molybdenum to the nontoxic molybdenum blue. The optimal pH, carbon source, molybdate concentration, and temperature supporting ...

We have evaluated three analytical methods for determining inorganic phosphorus in serum applied to the Technicon RA-I000 analyzer: a fully enzymatic colorimetric method based on the specific system purine nucleoside phosphorylase/xanthine oxidase coupled to an indicator colorimetric reaction similar to the Trinder reaction; a chemical method involving the direct UV measurement of the phosphomo...

The 5-day sodium carbonate-ammonium nitrate extraction assay (5-day method) has been recognized by the American Association of Plant Food Control Officials as a validated test method to identify fertilizers or beneficial substances that provide plant-available silicon (Si). The test method used the molybdenum blue colorimetric assay to quantify percentage Si; however, laboratories may use induc...

Molybdenum reduction to molybdenum blue by microbes is a potential bioremediation tool for molybdenum pollution. A previous work using cyanide as a respiratory inhibitor has shown that the site of molybdenum reduction in Enterobacter cloacae strain 48 is at the electron transport pathway. In the present work, several respiratory inhibitors together with cyanide were used to reinvestigate the si...

In ruminants, even trace amounts of molybdenum can be lethal. areas with high pollution, levels in soil and mine tailings exceed 20,000 ppm. Bioremediation challenging when toxic copper is also present. This research presents a novel approach using dialysis tubing the molybdenum-reducing activity Bacillus sp. strain Neni-8 for removal from aqueous solutions. Molybdenum blue (Mo-blue), produced ...
