Here we describe the expression pattern of the Na-K-2Cl-cotransporter NKKC1 during embryonal and early postnatal mouse development. During early stages hybridization signals were detected over single cells of the developing neuroepithelia, whereas the neuroepithelium of the basal telencephalon was labeled continuously. With ongoing differentiation a distinct pattern of hybridization became appa...

Stimulation of olfactory receptor neurons with odors culminates in opening of a ciliary Ca2+-activated Cl- channel. Because intracellular Cl- ([Cl-]i) is above electrochemical equilibrium in these cells, the result is cell depolarization that triggers action potentials that carry information to the olfactory bulb. In this issue of Neuron, Reisert and coworkers use combined pharmacological and m...

The branchial epithelium of the mudskipper Periophthalmodon schlosseri is densely packed with mitochondria-rich (MR) cells. This species of mudskipper is also able to eliminate ammonia against large inward gradients and to tolerate extremely high environmental ammonia concentrations. To test whether these branchial MR cells are the sites of active ammonia elimination, we used an immunological a...

The Rab GTPase-activating protein TBC1D4 (AS160) controls trafficking of the glucose transporter GLUT4 in adipocytes and skeletal muscle cells. TBC1D4 is also highly abundant in the renal distal tubule, although its role in this tubule is so far unknown. In vitro studies suggest that it is involved in the regulation of renal transporters and channels such as the epithelial sodium channel (ENaC)...

In the present report, we studied if an isoflavone, genistein, enhances the nerve growth factor (NGF)-induced neurite outgrowth of PC12 cells. Application of genistein enhanced the NGF-induced neurite outgrowth. Knockdown of Na+/K+/2Cl- cotransporter isoform 1 (NKCC1) abolished the stimulatory effect of genistein on the neurite outgrowth. These observations indicate that NKCC1 is essential for ...

A mathematical model of ascending Henle limb (AHL) epithelium has been fashioned using kinetic representations of Na+-K+-2Cl- cotransporter (NKCC2), KCC4, and type 3 Na+/H+ exchanger (NHE3), with transporter densities selected to yield the reabsorptive Na+ flux expected for rat tubules in vivo. Of necessity, this model predicts fluxes that are higher than those measured in vitro. The kinetics o...

In excitable cells, hypoxia inhibits K channels, causes membrane depolarization, and initiates complex adaptive mechanisms. It is unclear whether K channels of alveolar epithelial cells reveal a similar response to hypoxia. A549 cells were exposed to hypoxia during whole cell patch-clamp measurements. Hypoxia reversibly inhibited a voltage-dependent outward current, consistent with a K current,...
