Formobactin, a new free radical scavenger, was isolated from the culture of Nocardia sp. strain ND20. The structure of formobactin was determined to be a member of the nocobactin group antibiotics. Formobactin inhibited lipid peroxidation in rat brain homogenate. Formobactin also showed the activity to suppress L-glutamate toxicity in neuronal hybridoma N18-RE-105 cells.
Monascus purpureus is a red dye derived from yeast rice and has been used as color additives for food in East Asia. Monascus purpureus consists of several bioactive components. Some of these components work as a radical scavenger, thus monascus purpureus would also eliminate reactive oxygen species. Cancer cells maintain the high level of reactive oxygen species than normal cell and are death b...
Oxidative stress plays a pivotal role in developing and accelerating retinal diseases including age-related macular degeneration (AMD), glaucoma, diabetic retinopathy (DR), and retinal vein occlusion (RVO). An excess amount of reactive oxygen species (ROS) can lead to functional and morphological impairments in retinal pigment epithelium (RPE), endothelial cells, and retinal ganglion cells (RGC...
Purpose To assess the therapeutic potential of edaravone, a free radical scavenger that is used for the treatment of acute brain infarction and amyotrophic lateral sclerosis, in a mouse model of optic nerve injury (ONI). Methods Two microliters of edaravone (7.2 mM) or vehicle were injected intraocularly 3 minutes after ONI. Optical coherence tomography, retrograde labeling of retinal ganglio...
This study aimed to investigate the effects of mitochondrial ATP-sensitive potassium (MitoKATP) channel opening on the translocation of protein kinase C epsilon (PKCε). In addition, we aimed to determine the relationship between PKCε translocation and the production of reactive oxygen species (ROS). PKCε protein expression in cultured adult rat ventricular myocytes was investigated by immunoflu...
Relapse of disease and subsequent resistance to established therapies remain as major challenges in the treatment of multiple myeloma (MM). New therapeutic options are needed for these extensively pretreated patients. To explore an optimized combinational therapy, interactions between the irreversible proteasome inhibitor carfilzomib exhibiting a well-tolerated side-effect profile and histone d...
How the lens ages successfully is a lesson in biological adaption and the emergent properties of its complement of cells and proteins. This living tissue contains some of the oldest proteins in our bodies and yet they remain functional for decades, despite exposure to UV light, to reactive oxygen species and all the other hazards to protein function. This remarkable feat is achieved by a shrewd...
Insulin appears to play a role in brain physiology, and disturbances of cerebral insulin signalling and glucose homeostasis are implicated in brain pathology. The objective of the present study was to investigate the protective effects of insulin under conditions of oxidative stress induced by hydrogen peroxide (H2O2) in C6 glial cells. Insulin at concentration of 10−7 M could prevent 12 h H2O2...
