BC3H-1 myocytes were cultured in the presence of [3H]inositol or [3H]glucosamine during their entire growth cycle to ensure that all lipids containing inositol and glucosamine were labelled to isotopic equilibrium or maximal specific radioactivity. After such labelling, a lipid (or group of lipids), which was labelled with both inositol and glucosamine, was observed to migrate between phosphati...

Although ginsenoside Rg3 has been shown to exert anticancer effects on a variety of malignancies, the and molecular mechanisms oesophageal cancer remain unclear. Therefore, these factors must be explored. Here, we analyzed effect cell proliferation migration, as well its apoptosis. Furthermore, detected downstream pathways that could act on. The results showed significantly inhibited migration ...

The regulation of the synthesis of PtdIns(4,5)P2 is emerging as being as complex as we might expect from the multi-functional nature of this lipid. In the present chapter we focus on one aspect of inositide metabolism, which is the functions of the Type II PIPkins (Type II PtdInsP kinases). These are primarily PtdIns5P 4-kinases, although in vitro they will also phosphorylate PtdIns3P to PtdIns...

In eukaryotic cells, combinatorial phosphorylation of the hydroxyl residues on the inositol ring of phosphatidylinositol (PtdIns) gives rise to seven phosphoinositides (eight if PtdIns itself is included) (Fig. 1). In the ‘canonical’ phosphoinositide (PI) cycles, phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] serves as a precursor of the intracellular second The Physiology of Phosphoinos...

Fcgamma receptors mediate a variety of immune responses to IgG-containing complexes. Virtually all of these responses appear to be regulated by phosphoinositides, in particular phosphatidylinositol (4,5)P2 and phosphatidylinositol (3,4,5)P3. Possible downstream effectors of phosphoinositides involved in multiple FcgammaR-mediated events are discussed, as are elements of the signaling pathways t...

Resynthesis of phosphatidylinositol in permeabilized neutrophils following phospholipase Cβ activation : transport of the intermediate, phosphatidic acid, from the plasma membrane to the endoplasmic reticulum for phosphatidylinositol resynthesis is not dependent on soluble lipid carriers or vesicular transport Jacqueline WHATMORE*, Claudia WIEDEMANN*, Pennti SOMERHARJU†, Philip SWIGART* and Sha...

Phosphatidylinositol lipids, such as PI(4,5)P2 and PI(3,4,5)P3, are key mediators in diverse intracellular signaling pathways. Two recent reports examine how the metabolism of these lipids by phosphatidylinositol 3-kinases and the PTEN 3-phosphoinositide phosphatase may coordinate G protein coupled signaling pathways during eukaryotic chemotaxis.

The site of palmitoylation of the phosphatidylinositol moiety of the glycosyl-phosphatidylinositol membrane anchor of Trypanosoma brucei procyclic acidic repetitive protein was studied by using periodate oxidation. Analysis of the products by g.c.-m.s. allowed the assignment of 40 and 60% of the palmitate to the 2-position and the 3-position respectively of the myo-inositol ring.

Pollen tube cell volume changes rapidly in response to perturbation of the extracellular osmotic potential. This report shows that specific phospholipid signals are differentially stimulated or attenuated during osmotic perturbations. Hypo-osmotic stress induces rapid increases in phosphatidic acid (PA). This response occurs starting at the addition of 25% (v/v) water to the pollen tube culture...
