In cases of suspected extrapulmonary tuberculosis, rapid and accurate laboratory diagnosis is of prime importance, since traditional techniques of detecting acid-fast bacilli have limitations. The major difficulty with mycobacteria is achieving optimal cell lysis. Buffers used in commercial kits do not allow this complete lysis in a number of clinical specimens. A comparison of two sample prepa...

We report a comparative study of two DNA extraction techniques, thermolysis and chemical lysis (CTAB), for molecular identification and genotyping of M. tuberculosis. Forty DNA samples were subjected to PCR and the results demonstrated that with thermolysis it is possible to obtain useful data that enables fast identification and genotyping.

High quality DNA was isolated from Casuarina equisetifolia which is widely planted in tropics and subtropics for its multiple utilities. Two commonly used methods, CTAB based and the commercially supplied DNeasyTM Plant Mini Kit (QIAGEN GmbH, Germany) were tested for their efficiency in isolating high quantity and quality DNA to be utilized for AFLP analysis. The CTAB based protocol was modifie...

PCR is a highly accurate technique for confirming the presence of Mycobacterium avium subsp. paratuberculosis (Map) in broth culture. In this study, a simple, efficient, and low-cost method of harvesting DNA from Map cultured in liquid medium was developed. The proposed protocol (Universidad Austral de Chile [UACH]) was evaluated by comparing its performance to that of two traditional technique...

Current DNA extraction protocols, which require liquid nitrogen, lyophilization and considerable infrastructure in terms of instrumentation, often impede the application of biotechnological tools in less researched crops in laboratories in developing countries. We modified and optimized the existing CTAB method for plant genomic DNA extraction by avoiding liquid nitrogen usage and lyophilizatio...

Extraction of high quality RNA is a critical step in molecular genetics studies. Hazelnut is one of the most important nuts plants in the world. The presence of the taxol and other taxanes in hazelnut plant necessitates explaining their biosynthesis pathway and identifying the candidate genes. Therefore, an easy and practical method is necessary for RNA extraction from hazelnuts. Hazelnut has h...

A simple and efficient protocol for isolating genomic DNA from fresh and dry leaves of Vigna aconitifolia and V. trilobata was developed. It involves a modified CTAB procedure using 3% CTAB, 4% -mercaptoethanol, 2 M NaCl and 5% PVP. The extraction was carried out at 70°C. A slight increase in the concentrations of these chemical components and temperature helped in the removal of secondary meta...

A green, simple and sensitive spectrophotometric method was proposed for the determination of isoxsuprine hydrochloride (ISX), based on preconcentration with cetyltrimethylammonium bromide (CTAB) modified magnetic iron oxide nanoparticales (MNPs). ISX coupled diazotized metoclopramide drug via diazotization-coupling reaction in alkaline medium to form yellow-orange color product which extracted...
