This work reports endoglucanase and beta-glucosidase production by the thermophilic fungus Myceliophthora heterothallica in solid-state (SSC) and submerged (SmC) cultivation. Wheat bran and sugarcane bagasse were used for SSC and cardboard for SmC. Highest endoglucanase production in SSC occurred after 192 hours: 1,170.6 ± 0.8 U/g, and in SmC after 168 hours: 2,642 ± 561 U/g. The endoglucanases...

α-Glucosidases I and II are endoplasmic reticulum-resident enzymes that are essential for N-linked glycan processing and subsequent proper folding of glycoproteins. In this report, we first demonstrate that downregulation of the expression of α-glucosidase I, II, or both in Huh7.5 cells by small hairpin RNA technology inhibited the production of hepatitis C virus (HCV). In agreement with the es...

Isolation of novel thermophilic bacilli has received considerable attention among the scientific community in whole world because of their biotechnological importance as they possess unique enzymes with thermal activity and stability. Of the thermophilic enzymes, we were interested in α-glucosidases which catalyze hydrolysis of terminal non-reducing α-D-glucosidic linkages of oligoand polysacch...

1. The behaviour of rat liver alpha-glucosidases on dextran gel (Sephadex G-100) columns was studied. A ;retardation' of an acid alpha-glucosidase activity was observed. This activity was identified as lysosome alpha-(1-->4)-glucosidase. A single gel-filtration step resulted in a 700-fold purification of the enzyme. The same technique was also used to purify the acid alpha-glucosidase of human ...

β-Glucosidases (β-D-glucoside glucohydrolases) play important role in nature, including the degradation of cellulosic biomass by fungi and bacteria, breakdown of glycolipids in mammalian lysosomes, and the cleavage of glycosylated flavonoids in plants. These enzymes have broad substrate specificity, and are used in a range of biotechnological processes. The most intensively studied area of thei...

BACKGROUND A complete saccharification of plant polymers is the critical step in the efficient production of bio-alcohols. Beta-glucosidases acting in the degradation of intermediate gluco-oligosaccharides produced by cellulases limit the yield of the final product. RESULTS In the present work, we have identified and then successfully cloned, expressed, purified and characterised 4 highly act...

Starch digestion in the human body is typically viewed in a sequential manner beginning with α-amylase and followed by α-glucosidase to produce glucose. This report indicates that the two enzyme types can act synergistically to digest granular starch structure. The aim of this study was to investigate how the mucosal α-glucosidases act with α-amylase to digest granular starch. Two types of enzy...

the rose sawfly, arge rosae linnaeus, is one of the most destructive pests of rose bushes in the north of iran. nowadays, many attempts have been made to reduce pesticide application by looking for new methods of pest control. a non chemical method for controlling insect pests including a. rosae can be achieved by using genetically engineered plants expressing carbohydrase inhibitors. therefore...
