The synthesis and characterisation, including solid state structure, of a macrocycle containing both a hydrazone and a disulfide linkage is described. Selective ring-opening of the macrocycle under thermodynamic control could be achieved at either the disulfide or the hydrazone linkage by applying mutually exclusive sets of reaction conditions.

The asymmetric unit of the title compound, C30H46N6O4, contains one half-mol-ecule. The C(benzene)-C(CH2)-N-C(-Me) torsion angle is -79.89 (13)° suggesting a synclinal orientation of the nitro-benzene ring with respect to the macrocycle. The conformation of the macrocycle is stabilized by intra-molecular N-H⋯N hydrogen bonds.

In this report, we present cyclohexylcyanine, a phthalocyanine analogue with two cyclohexane units incorporated into the backbone of the macrocycle; in the free base, the ionizable protons lie on the periphery of the ring, and when metalated with silver the macrocycle binds to the metal via these same meso nitrogen atoms.

We report cystine-based macrocyclic compounds showing very unique self-assembling behavior. 24-membered disulfide macrocycle 3 shows a helical and tubular organization and also forms an insoluble plaque that is birefringent with congo red dye. 48-membered macrocycle 4 shows a vesicle-like assembly with capability for encapsulation, as demonstrated by rhodamine B encapsulation.

A metallosupramolecular [Pd2L4](4+) cage can be cleanly converted into a [Pd2L2Cl4] metallo-macrocycle upon addition of chloride ions. The process is reversible, treatment of the [Pd2L2Cl4] macrocycle with silver(I) ions regenerates the [Pd2L4](4+) cage. Additionally, it is shown that guest molecules could be released on chloride triggered cage dis-assembly and taken up anew on re-assembly.

The preparation of a Syringolin A/Glidobactin A hybrid (SylA-GlbA) consisting of a SylA macrocycle connected to the GlbA side chain and its potent proteasome targeting of all three proteasomal subsites is reported. A rotaxane incorporating three different stations and fluorescent states (output) was prepared. The movement of the macrocycle can be easily detected by fluorescence change as an output signal and the macrocycle could be easily controlled to locate on three different stations of the thread by the tuning of acid/base (input).
