We report here the ~670 Mb genome assembly of the Asian seabass (Lates calcarifer), a tropical marine teleost. We used long-read sequencing augmented by transcriptomics, optical and genetic mapping along with shared synteny from closely related fish species to derive a chromosome-level assembly with a contig N50 size over 1 Mb and scaffold N50 size over 25 Mb that span ~90% of the genome. The p...

OBJECTIVE To investigate possible altered CNS excitability in vaginismus. METHODS In 10 patients with primary idiopathic lifelong vaginismus, 10 with vulvar vestibulitis syndrome accompanied by vaginismus and healthy controls we recorded EMG activity from the levator ani (LA) and external anal sphincter (EAS) muscles and tested bulbocavernosus reflex (BCR). Pudendal-nerve somatosensory evoked...

Efficient sequencing of animal and plant genomes by next-generation technology should allow many neglected organisms of biological and medical importance to be better understood. As a test case, we have assembled a draft genome of Caenorhabditis sp. 3 PS1010 through a combination of direct sequencing and scaffolding with RNA-seq. We first sequenced genomic DNA and mixed-stage cDNA using paired ...

A field experiment was conducted during kharif season of 2010 and 2011 to investigate the optimization of nitrogen levels under different age of seedlings transplanted on growth, chlorophyll content, yield and economics of hybrid rice. Fifteen treatment combinations consisted of three levels of seedlings age (10, 20 and 30 days) and five levels of nitrogen viz. N0, N50, N100, N150 and N200 kg h...

We previously described the whole-genome assembly program Arachne, presenting assemblies of simulated data for small to mid-sized genomes. Here we describe algorithmic adaptations to the program, allowing for assembly of mammalian-size genomes, and also improving the assembly of smaller genomes. Three principal changes were simultaneously made and applied to the assembly of the mouse genome, du...

Butterflies are exceptionally diverse but their potential as an experimental system has been limited by the difficulty of deciphering heterozygous genomes and a lack of genetic manipulation technology. Here we use a hybrid assembly approach to construct high-quality reference genomes for Papilio xuthus (contig and scaffold N50: 492 kb, 3.4 Mb) and Papilio machaon (contig and scaffold N50: 81 kb...

recently, biological fertilizers extensively recommended because of over application of chemical fertilizer and their effects on environment and production resources. in order to study the effect of inoculate phosphate solubilization microorganisms (psm) and plant growth promoting rhizobacteria (pgpr) on nutrient use efficiency in corn [zea mays (l.) cv. sc 604] an experiment was conducted in 2...

We describe documented simultaneous intracerebral auditory evoked potentials from the auditory cortex and medial geniculate body (MGB) of a human patient. The MGB response lasted > 300 ms, with an initial negativity at 13.5 ms (N13), two positive peaks P21 and P29, and two broader negativities N50 and N200. P21 and N50 amplitudes were strongest for lowest tone frequencies, suggesting possible M...

We have developed a new set of algorithms, collectively called "Velvet," to manipulate de Bruijn graphs for genomic sequence assembly. A de Bruijn graph is a compact representation based on short words (k-mers) that is ideal for high coverage, very short read (25-50 bp) data sets. Applying Velvet to very short reads and paired-ends information only, one can produce contigs of significant length...
