A key stage in determining the phenotype(s) conferred by a plasmid is its displacement, or 'curing,' to create a plasmid-free strain. However, many plasmids are very stable, not only because they contain multiple replicons, but also because they can encode post-segregational killing systems that reduce the viability of plasmid-free segregants. We have developed an efficient curing strategy that...

The potentially probiotic strain Lactobacillus plantarum CCUG 43738, which displayed atypical phenotypic resistance to tetracycline (MIC, 512 microg/ml) and minocycline (MIC, 256 microg/ml), was found to contain a tet(S) gene located on a plasmid of approximately 14 kb. Plasmid curing with novobiocin eliminated this plasmid and restored the tetracycline-susceptible phenotype of the host strain.

A study was conducted to determine plasmid mediated methicillin and vancomycin resistant Staphylococcus aureus along with plasmid profiling and detection of vancomycin resistance genes. This study was carried out in the Department of Microbiology and Fermentation Technology, Sam Higginbottom Institute of Agriculture Technology and Sciences, Allahabad, India. The molecular detection of gene was ...

Exposure to bile induces curing of the virulence plasmid in Salmonella enterica serovar Typhimurium (pSLT). Disruption of the ccdB gene increases pSLT curing, both spontaneous and induced by bile, suggesting that the pSLT ccdAB genes may encode a homolog of the CcdAB addiction module previously described in the F sex factor. Unlike the F sex factor, synthesis of pSLT-encoded pili does not confe...

As the causative agent of foodborne related illness, Vibrio species causes a huge impact on the public health and management. Vibrio species is often associated with seafood as the latter plays a role as a vehicle to transmit bacterial infections. Hence, antibiotics are used not to promote growth but rather to prevent and treat bacterial infections. The extensive use of antibiotics in the aquac...

Agrobacterium tumefaciens harboring a Ti plasmid causes plant tumorigenesis and it's DNA transfer system have been extensively used in plant biotechnology. Because Ti plasmids are high stable in agrobacterial hosts, their curing is very difficult by conventional methods. Here, we introduced a novel curing method by using Ti plasmids incompatibility.

A strain of Staphylococcus epidermidis was transduced to tobramycin resistance and all transductants were also resistant to kanamycin and neomycin. Results of curing studies also indicated that these resistances were controlled by a single determinant on a plasmid. Agarose gel electrophoresis of the plasmid DNA from parent, cured, and transduced strains showed a single plasmid was responsible a...

Curing of the P1 plasmid prophage in recA(P1) lysogens by superinfection with another P1 phage was specific and independent of immunity and incompatibility expression.
