Optimal enzyme mixtures of six Trichoderma reesei enzymes and five thermostable enzyme components were developed for the hydrolysis of hydrothermally pretreated wheat straw, alkaline oxidised sugar cane bagasse and steam-exploded bagasse by statistically designed experiments. Preliminary studies to narrow down the optimization parameters showed that a cellobiohydrolase/endoglucanase (CBH/EG) ra...
Dilute sulfuric acid (DA), sulfur dioxide (SO(2)), liquid hot water (LHW), soaking in aqueous ammonia (SAA), ammonia fiber expansion (AFEX), and lime pretreatments were applied to Alamo, Dacotah, and Shawnee switchgrass. Application of the same analytical methods and material balance approaches facilitated meaningful comparisons of glucose and xylose yields from combined pretreatment and enzyma...
Abstract Background Filamentous fungus Trichoderma reesei has been widely used as a workhorse for cellulase and xylanase productions. Xylanase reported the crucial accessory enzyme in degradation of lignocellulose higher accessibility cellulase. In addition, efficient hydrolysis xylan needs co-work multiple xylanolytic enzymes, which rise an increasing demand high yield biomass degradation. Res...
Rhizopus stolonifer was cultivated in wheat bran to produce a cellulase-free alkaline xylanase. The purified enzyme obtained after molecular exclusion chromatography in Sephacryl S-200 HR showed optimum temperature as 45o C and hydrolysis pHs optima as pH 6.0 and 9.0. Xylanase presented higher Vmax at pH 9.0 (0.87 μmol/mg protein) than at pH 6.0 and minor Km at pH 6.0 (7.42 mg/mL) than at pH 9.0.
Aeromonas caviae W-61 produces multiple extracellular xylanases, the xylanases 1, 2, 3, 4, and 5. In this study, we purified and characterized the xylanase 5 of A. caviae W-61, and amplified a part of xylanase 5 gene (xyn5). The purified xylanase 5 was found to be a single polypeptide with molecular mass of 140 kDa. It was an endo-beta-1,4-xylanase showing optimum temperature 40 degrees C and o...
A xylan-degrading enzyme (xylanase II) was purified to apparent homogeneity from solid-state cultures of Aspergillus fumigatus Fresenius. The molecular weight of xylanase II was found to be 19 and 8.5 kDa, as estimated by SDS-PAGE and gel filtration on FPLC, respectively. The purified enzyme was most active at 55C and pH 5.5. It was specific to xylan. The apparent K m and V max values on solubl...
BACKGROUND In recent years, xylanases have attracted considerable research interest because of their potential in various industrial applications. The yeast Pichia pastoris can neither utilize nor degrade xylan, but it possesses many attributes that render it an attractive host for the expression and production of industrial enzymes. RESULTS The Xyn2 gene, which encodes the main Trichoderma r...
For producing second-generation ethanol (cellulosic ethanol) and other value-added bioproducts, magnetic graphene oxide (GO-MNP) was synthesized in this work used as the immobilization support for an industrial cellulase-and xylanase-containing preparation. GO-MNP characterization by TEM, SEM ATR-FTIR spectroscopy showed that nanoparticles are homogeneously distributed onto GO sheets surface. T...
background and objectives: bacillus species are attractive industrial organisms due to their rapid growth rates leading to a short fermentation cycle and for their capacity to secrete important enzymes and proteins such as xylanase into the extracellular medium. considering the industrial importance of xylanase, in this current study, bacillus spp. were isolated from different soils and were sc...
Hydrolysis of xylan, the chief type of hemicellulose is achieved by endo-1, 4-β-xylanase and βxylosidase among other such enzyme complex. These enzymes are mainly produced by fungi, bacteria, etc. Xylanase finds applications in animal feed, manufacture of bread, beverages, textiles, bleaching cellulose pulp, ethanol and xylitol production. Xylanase depolymerizes xylan molecules into xylose unit...
