Peptide analogues targeting various neuropeptide receptors have been used effectively in cancer therapy. A hallmark of adrenocortical tumor formation is the aberrant expression of peptide receptors relating to uncontrolled cell proliferation and hormone overproduction. Our microarray results have also demonstrated a differential expression of neuropeptide hormone receptors in tumor subtypes of ...

Treatment with the NMDA receptor antagonist MK-801 prevented the adrenal steroid-induced suppression of cell death, determined by both morphological identification of pyknotic cells and TUNEL staining, in the dentate gyrus in rat pups. This finding suggests that adrenal steroids naturally promote granule cell survival via NMDA receptor activation.

BACKGROUND Adenomas of the adrenal gland are rare causes of virilization in childhood. CASE REPORT A girl aged 2 years and 4 months presented with pubarche, distinct clitoral hypertrophy, tall stature, and increased height velocity. Plasma testosterone and dehydroepiandrosterone were elevated. Androgens remained unchanged after adrenocorticotropic hormone, and dexamethasone administrations. U...

Seizures, including electrically induced seizures (ECS), activate the hypothalamo-pituitary-adrenal (HPA) axis in rats. The present studies were undertaken to characterize the effects of repeated ECS on hormone release and brain adrenal steroid receptors. Repeated ECS led to an increase in adrenal weight, an increase in the corticosterone response to the eighth seizure compared to the first sei...

The nuclear receptor steroidogenic factor 1 (SF-1) is essential for adrenal development and steroidogenesis. The atypical orphan nuclear receptor Dax-1 binds to SF-1 and represses SF-1 target genes. Paradoxically, however, loss-of-function mutations of Dax-1 also cause adrenal hypoplasia, suggesting that Dax-1 may function as an SF-1 coactivator under some circumstances. Indeed, we found that D...

Expression of tyrosine receptor kinase B (TrkB), a receptor for brain-derived neurotrophic factor (BDNF), is markedly elevated in the adrenal medulla during immobilization stress. Catecholamine release was confirmed in vitro by stimulating chromaffin cells with recombinant BDNF. We investigated the role of TrkB and the localization of BDNF in the adrenal gland during immobilization stress for 6...

Patients with an aldosterone-producing adenoma (APA) characteristically fail to show an increase in plasma aldosterone (PA) concentration with maneuvers that increase angiotensin II (Ang II), yet they retain a brisk response of PA to adrenocorticotrophic hormone. Therefore, adrenal Ang II receptor binding was characterized in a patient with APA who had a blocked PA response to Ang II infusion b...

The contribution of neuropeptide Y (NPY), deriving from adrenal medulla, to the adrenosympathetic tone is unknown. We found that in response to NPY, primary cultures of mouse adrenal chromaffin cells secreted catecholamine, and that this effect was abolished in cultures from NPY Y(1) receptor knockout mice (Y(1)-/-). Compared with wild-type mice (Y(1)+/+), the adrenal content and constitutive r...

Regulation of tyrosine hydroxylase (TH) enzymatic activity in vivo by muscarinic receptor agonists in rat adrenal medulla was characterized in this study. Bethanechol and carbachol produce dose-dependent increases in rat adrenal TH activity. These increases are maximal (approximately 3-fold) using 10 mg/kg bethanechol or 0. 5 mg/kg carbachol and are totally inhibited by prior administration of ...

In the present work, the presence of gastric inhibitory polypeptide (GIP) receptors and their functional role in the adrenal cells of three patients with food-dependent Cushing's syndrome were studied. RT-PCR and in situ hybridization studies demonstrated the presence of GIP receptor in the adrenals of the three patients. The presence of this receptor was also demonstrated in two human fetal ad...
