We report a new strategy for cloning DNA of the plant-infecting geminiviruses that is based on PCR amplification with primers overlapping the sequence of a restriction site in the viral genome. This method reduces the time and work necessary to obtain biologically active geminivirus DNAs and could be used for other DNA viruses having circular genomes, such as the animalinfecting circoviruses. G...

Geminiviruses are small DNA viruses that use plant replication machinery to amplify their genomes. Microarray analysis of the Arabidopsis (Arabidopsis thaliana) transcriptome in response to cabbage leaf curl virus (CaLCuV) infection uncovered 5,365 genes (false discovery rate <0.005) differentially expressed in infected rosette leaves at 12 d postinoculation. Data mining revealed that CaLCuV tr...

DNA methylation is a fundamental epigenetic modification that regulates gene expression and represses endogenous transposons and invading DNA viruses. As a counter-defense, the geminiviruses encode proteins that inhibit methylation and transcriptional gene silencing (TGS). Some geminiviruses have acquired a betasatellite called DNA β. This study presents evidence that suppression of methylation...

Geminiviruses are single-stranded circular DNA viruses causing leaf curl disease in papaya crop. Post-Transcriptional Gene Silencing (PTGS), also known as RNAi, acts as a natural antiviral defence mechanism and plays a role in genome maintenance and development in plants. PTGS suppression by viruses makes the plant RNA silencing machinery inefficient. Three geminiviral genes namely AV2, AC2 and...

Plant viral movement proteins mediate the cell-to-cell movement of nucleic acids. This involves either a direct interaction between the viral movement protein and the nucleic acid or an indirect interaction involving host factors. The bipartite geminiviruses possess two movement proteins, BV1 and BC1, that coordinate movement of viral DNA across nuclear and plasmodesmal boundaries, respectively...

Implementation of a vector-enabled metagenomics approach resulted in the identification of various geminiviruses. We identified the genome sequences of Beet curly top Iran virus, Turnip curly top viruses, Oat dwarf viruses, the first from Iran, and Wheat dwarf virus from leafhoppers feeding on beet, parsley, pumpkin, and turnip plants.
