Protocol: To detect the 506 polymorphism, a 567 bp SMPD1 genomic fragment is amplified using sense (5'-AGTAGTCGACATGGGCAGGATGTGTGG-3') and antisense (5'-AGTAGTGTCGACTTGCCTGGTTGAACC AC AGC-3') primers. Dot-blot hybridization is performed using allele-specific oligonucleotides for 506-Arg (5'-ACTACTCCAGGAGCTCT-3') and for 506-Gly (5'-ACTACTCCGGGAGCTCT-3'), which are hybridized at 42°C and washed ...
Empirical data sets of intraspecific restriction site polymorphism in Drosophila have been gathered in order to test hypotheses derived from coalescent theory. Three main ideas are tested: (1) haplotype frequency in the sample contains information on the topological position of a given haplotype in a cladogram, (2) the frequency of a haplotype is related to the number of mutational connections ...
The wealth of sequence information available for Neurospora crassa and other fungi has greatly facilitated evolutionary and molecular analyses of this group. Although "reverse" genetics, in which genes are first identified by their sequence rather than by their mutant phenotypes, serves as a valuable new approach for elucidating biological processes, classical "forward" genetic analysis is stil...
Due to the size of BAC, PAC and P1 clones, it is often difficult to construct detailed restriction maps, with large number of restriction fragments leading to ambiguity of mapping data. We report the use of Cre recombinase to linearise and asymmetrically introduce label at the unique loxP site of large loxP-containing clones. Subsequent partial digestion allows the direct ordering of restrictio...
A set of 147 Dictyostelium discoideum strains was constructed by random integration of a vector containing rare restriction sites. The strains were generated by transformation using restriction enzyme-mediated integration (REMI) which results in the integration of linear DNA fragments into randomly distributed genomic restriction sites. Restriction fragment length polymorphism (RFLP) was genera...
MOTIVATION To determine the most powerful artificial intelligence techniques for automated restriction mapping, and use them to create a powerful multiple-enzyme restriction mapping tool. RESULTS The most effective search engine utilized model-driven exhaustive search and a form of binary logic pruning based on Pratt's separation theory. Additional experimentation led to the development of an...
From a genomic library constructed from sperm DNA of the inbred BALB/c mouse, we previously isolated 54 cosmid clones that contain 36 class I genes and can be divided by restriction map analyses into 13 gene clusters. We have isolated single- and low-copy DNA probes from each of these clusters to visualize restriction enzyme site polymorphisms in the DNAs from various congeneic and recombinant ...
where f(rea) =f(z) is any analytic function regular in a circle of radius R>r about the origin. We can define a particular complex valued random variable Z in the following way: \Z\ =r with probability one, and arg Z is uniformly distributed on the interval [— x, x]. We shall denote by E the linear operator which "takes the expected value" of Z and of measurable functions of Z with respect to t...
1.1. Sheaves. Let X be a topological space. A sheaf F on X associates to each open set U an abelian group F (U), called the sections of F over U , along with a restriction map rV,U : F (V ) → F (U) for any open sets U ⊂ V (for a section σ ∈ F (V ), we often write σ|U to denote rV,U (σ)), satisfying the following conditions: (1) For any open sets U ⊂ V ⊂W , rV,U ◦ rW,V = rW,U ; (2) For a collect...
We show the dominance of the restriction map from a moduli space of stable sheaves on the projective plane to the Coble sixfold quartic. With the dominance and the interpretation of a stable sheaf on the plane in terms of hyperplane arrangements, we expect these tools to reveal the geometry of the Coble quartic.
