Rat liver nuclear protein kinase NI, which appears in the flowthrough of DEAE-Sephadex columns, has been purified approximately 15,000-fold from soluble nuclear protein with yields of up to 10%. The method of purification involved chromatography of the DEAE-flowthrough protein successively on phosvitin-Sepharose and casein-Sepharose followed by rechromatography on phosvitin-Sepharose. The purif...

Desulfurivibrio alkaliphilus strain AHT2(T) is a strictly anaerobic sulfidogenic haloalkaliphile isolated from a composite sediment sample of eight hypersaline alkaline lakes in the Wadi al Natrun valley in the Egyptian Libyan Desert. D. alkaliphilus AHT2(T) is Gram-negative and belongs to the family Desulfobulbaceae within the Deltaproteobacteria. Here we report its genome sequence, which cont...

Community composition of the infaunal bivalve fauna of the St. Lucie Estuary and southern Indian River Lagoon, eastern Florida was sampled quarterly for 10 years as part of a long-term benthic monitoring program. A total of 38,514 bivalves of 137 taxa were collected and identified. We utilized this data, along with sediment samples and environmental measurements gathered concurrently, to assess...

A mixing model derived from first principles describes the bulk density (BD) of intertidal wetland sediments as a function of loss on ignition (LOI). The model assumes that the bulk volume of sediment equates to the sum of self-packing volumes of organic and mineral components or BD = 1/[LOI/k1 + (1-LOI)/k2], where k1 and k2 are the self-packing densities of the pure organic and inorganic compo...

BACKGROUND Pretreatment is a vital but expensive step in biomass biofuel production. Overall, most of this past effort has been directed at maximizing sugar yields from hemicellulose and cellulose through trials with different chemicals, operating conditions, and equipment configurations. Flowthrough pretreatment provides a promising platform to dissolution of lignocellulosic biomass to generat...

UNLABELLED Two acid mine drainage (AMD) sites in the Appalachian bituminous coal basin were selected to enrich for Fe(II)-oxidizing microbes and measure rates of low-pH Fe(II) oxidation in chemostatic bioreactors. Microbial communities were enriched for 74 to 128 days in fed-batch mode, then switched to flowthrough mode (additional 52 to 138 d) to measure rates of Fe(II) oxidation as a function...

Magnetic pore fabrics (MPFs) serve as efficient proxy for alignment and preferential flow directions. Anisotropy of magnetic susceptibility impregnated samples reflects the shape arrangement ferrofluid-filled pores. Reliable interpretation requires that entire connected space is reached by ferrofluid. This study investigates impregnation efficiencies various methods (standard vacuum, pressure i...

The initial benthic degradation of senescent Zostera marina leaves was studied in controlled flowthrough microcosm chambers for 33 d. Sediment chambers without added eelgrass leaves served as control chambers. The inflowing artificial seawater and ouflowing seawater were analyzed for dissolved organic carbon and nitrogen (DOC and DON), total acid-hydrolyzable amino acids (THAA), dissolved free ...

We orally inoculated Rana catesbeiana tadpoles (n=23) and metamorphs (n=24) to test their suitability as hosts for Escherichia coli O157:H7. Tadpoles were housed in flowthrough aquaria and did not become infected. Metamorphs were housed in stagnant aquaria, and 54% tested positive through 14 days postinoculation, suggesting that they are suitable hosts for E. coli O157:H7.
