Protoplast wet densities (1.315 to 1.400 g/ml), determined by buoyant density sedimentation in Metrizamide gradients, were correlated inversely with the protoplast water contents (26.4 to 55.0 g of water/100 g of wet protoplast) of nine diverse types of pure lysozyme-sensitive dormant bacterial spores. The correlation equation provided a precise method for obtaining the protoplast water content...
The fresh-water crustacean Daphnia magna may acquire an infection with the microsporidium Octosporea bayeri either by ingesting spores from the water (horizontally), or directly from its mother (vertically). Due to differences in the time and mechanisms of transmission, horizontal and vertical infections may lead to differences in the growth of the parasite within the host. This may influence p...
Phytoextraction involves use of plants to remove toxic metals from soil. We examined the effects of phytoextraction practices with three plant species (Silene vulgaris, Thlaspi caerulescens, and Zea mays) and a factorial variation of soil amendments (either an ammonium or nitrate source of nitrogen and the presence or absence of an elemental sulfur supplement) on arbuscular mycorrhizal (AM) fun...
Spore formation in the Gram-positive bacterium Bacillus subtilis has been classically viewed as an example of unicellular differentiation that occurs in response to nutritional starvation. We present evidence that B. subtilis produces an extracellular factor(s) that is required, in addition to starvation conditions, for efficient sporulation. This factor is secreted and accumulates in a cell de...
Reduction of resting spore density ofPlasmodiophora brassicaeand clubroot disease severity by liming
Bacterial spores remain dormant and highly resistant to environmental stress until they germinate. Completion of germination requires the degradation of spore cortex peptidoglycan by germination-specific lytic enzymes (GSLEs). Bacillus anthracis has four GSLEs: CwlJ1, CwlJ2, SleB, and SleL. In this study, the cooperative action of all four GSLEs in vivo was investigated by combining in-frame de...
Penicillin-binding proteins (PBPs) are enzymes involved in the synthesis of peptidoglycan structures in Bacillus subtilis such as the vegetative cell wall and the spore cortex. The B. subtilis sequencing project has identified a gene (orf16, EMBL accession number D38161) which exhibits significant sequence similarity to genes encoding class B high-molecular-weight PBPs. We have found that orf16...
The low heat resistance (D100 = 0.554 min, z = 13.4 degrees C) of dormant lysozyme-sensitized spores of Bacillus sphaericus 9602 was correlated with a low protoplast wet density (1.305 g/ml) equivalent to a high protoplast water content (61.0%, wet weight basis). These values for these unusual spores were consistent with those correlated previously in 28 spore types of seven other species.
Crop plants genetically modified for the expression of Bacillus thuringiensis (Bt) insecticidal toxins have broad appeal for reducing insect damage in agricultural systems, yet questions remain about the impact of Bt plants on symbiotic soil organisms. Here, arbuscular mycorrhizal fungal (AMF) colonization of transgenic maize isoline Bt 11 (expressing Cry1Ab) and its non-Bt parental line (Provi...
Root distribution and mycorrhizal associations were compared in primary, secondary and limestone forests in Xishuangbanna, southwest China. Soil cores to a depth of 20 cm were collected at random points from four 50 m2 quadrats in each forest type. Arbuscular mycorrhizal (AM) associations were the only form of mycorrhiza found in all forest types. The primary forest was characterized by high ro...
