Gastrointestinal (GI) anthrax results from the ingestion of Bacillus anthracis. Herein, we investigated the pathogenesis of GI anthrax in animals orally infected with toxigenic non-encapsulated B. anthracis Sterne strain (pXO1+ pXO2-) spores that resulted in rapid animal death. B. anthracis Sterne induced significant breakdown of intestinal barrier function and led to gut dysbiosis, resulting i...

for evaluation the stability and potency of anthrax vaccine prepared in razi vaccine and serum research institute in iran, samples of different batches of vaccine were kept at 4-8 °c (refrigerator), 20-25 °c (room temperature) and 37 °c (incubator).the viable spores/ml of vaccines were determined using plate–counting method before and after holding at different temperatures monthly. vaccine pot...

Recent bioterrorism concerns have prompted renewed efforts towards understanding the biology of bacterial spore resistance to radiation with a special emphasis on the spores of Bacillus anthracis. A review of the literature revealed that B. anthracis Sterne spores may be three to four times more resistant to 254-nm-wavelength UV than are spores of commonly used indicator strains of Bacillus sub...

Bacillus anthracis Sterne cured of the pXO1 plasmid had enhanced secreted protease activity during the postexponential phase but no change in hemolytic or lecithinase activities. A zymogen profile revealed at least six proteases, including serine, metal, and perhaps cysteine types. There were similar amounts of protease secreted by the closely related species Bacillus cereus and Bacillus thurin...

Improved culture conditions and a new, completely synthetic medium (R medium) were developed to facilitate the production of Bacillus anthracis holotoxin antigens. Levels of these antigens up to fivefold greater than the highest previously reported values were recovered with the described system. Cultures of Sterne, V770-NP1-R, and Vollum 1B strains of B. anthracis were monitored for growth, pH...

The Sterne and Ames strains of Bacillus anthracis carry chromosomal genes bla1 and bla2, which confer beta-lactam resistance when expressed in Escherichia coli. MIC measurements and steady-state kinetic analyses indicate that Bla1 possesses penicillinase activity while Bla2 possesses penicillinase, cephalosporinase, and carbapenem-hydrolyzing activities.

Bacillus anthracis is a National Institute of Allergy and Infectious Diseases Category A priority pathogen and the causative agent of the deadly disease anthrax. We applied a transposon mutagenesis system to screen for novel chromosomally encoded B. anthracis virulence factors. This approach identified ClpX, the regulatory ATPase subunit of the ClpXP protease, as essential for both the hemolyti...

Bacillus anthracis, the causative agent of anthrax, relies on multiple virulence factors to subvert the host immune defense. Using Caenorhabditis elegans as an infection model, we screened approximately 5,000 transposon mutants of B. anthracis Sterne for decreased virulence. One of the attenuated mutants resulted in loss of expression of yceG and yceH, the last two genes in a six-gene cluster o...

Distressing effects on animal and human health with lethal progression, being used as bioweapon shared features non-pathogenic bacteria demands sensitive, specific, safe, cost effective rapid detection methods for anthrax causing organisms. Conventional microbiology based diagnostics are time consuming need sophisticated equipment, while molecular require less labor. The Loop mediated isotherma...

Bacterial spores are encased in a multilayered proteinaceous shell, called the coat. In many Bacillus spp., the coat protects against environmental assault and facilitates germination. In Bacillus anthracis, the spore is the etiological agent of anthrax, and the functions of the coat likely contribute to virulence. Here, we characterize a B. anthracis spore protein, called Cotbeta, which is enc...
